Abstract

Namen diplomskega dela je bila validacija analizne metode za določanje glukozinolatov v semenih navadnega rička. Določili smo vsebnosti glukozinolatov z visokotlačno tekočinsko kromatografijo (po dveh različnih metodah) v 27 vzorcih slovenskega porekla, treh zaporednih letin (2007,2008 in 2009). Za primerjavo smo določili glukozinolate še 4 vzorcem iz tujine (Danska, Nemčija, ZDA in Finska). Vzorcem smo določili še delež vlage in količino olja. Opravili smo validacijo analizne metode, določili linearnost, natančnost, mejo določljivosti, časovno stabilnost ekstrakta in vpliv vremenskih razmer na vsebnost glukozinolatov. Na osnovi vseh rezultatov smo ugotovili, da gre v Sloveniji za eno samo vrsto navadnega rička, saj se nikjer med analiziranimi vzorci ne pojavljajo signifikantne razlike.

Secondary title: Validation of analytical method for determination of glucosinolates in false flax seeds (Camelina sativa)
Secondary abstract: Purpose of this thesis was the validation of the analytical method for the determination of glucosinolates in the seeds of false flax. We have determined the content of glucosinolates with high pressure liquid chromatography, using two different methods. We have used 27 samples of Slovenian origin from three consecutive harvests (2007, 2008 and 2009). For comparison, we have determined the glucosinolates content in 4 other samples. These samples originated from Denmark, Germany, USA and Finland. Furthermore, we have also determined the moisture content and the quantity of oil. We have performed complete validation of the analytical method. We have determined linearity, precision, limit of quantification, temporal stability of the extract and the impact of weather on the content of glucosinolates. Based on all final results, we have concluded that in Slovenia exists only one species of false flax, because there were no significant differences between the analyzed samples.
