Abstract

Cilj diplomske naloge je primerjava rezultatov koncentracij C-reaktivnega proteina z dvema različnima rutinskima metodama nefelometrično in turbidimetrično. Uporabljeni vzorci so serumi, dobljeni iz krvi pacientov v Splošni bolnišnici dr. Jožeta Potrča na Ptuju. Analizirali smo 90 vzorcev C-reaktivnega proteina (CRP) in 90 vzorcev C-reaktivnega proteina visoke občutljivosti (hs-CRP). Vsak vzorec smo analizirali na dveh različnih instrumentih, nefelometričnem analizatorju BN ProSpec in spektrofotometričnem analizatorju Dimensions RxL Max. Priozvajalec instrumentov je bil za oba enak Dade Behring. Iz statističnega ovrednotenja rezultatov lahko zaključimo, da je nefelometrična metoda bolj natančna, ker je metoda bolj občutljiva in specifična. Iz rezultatov linearnih regresijskih koeficientov, Passing Bablokove regresijske premice in iz rezultatov parametričnih statističnih testov lahko zaključimo, da turbidimetrično metodo lahko zamenjamo z nefelometrično metodo in da sta metodi medsebojno primerljivi.

Secondary abstract: Aim of diploma thesis is to compare the results of C-reactive protein concentrations in different samples using two different routine methods nephelometric and turbidimetric. The samples are serums, obtained from blood of patients in General Hospital dr. Jože Potrč in Ptuj. 90 samples C-reactive protein (CRP) and 90 samples of C-reactive protein high sensitivity (hs-CRP) are analysed. Each sample was analysed by two methods at two different instruments, namely nephelometric analyzer BN ProSpec and spectrophotometric analyzer Dimensions RxL Max. The instruments are purchased by the same manufacturer, Dade Behring. From the statistical evaluation of measurement results, we concluded that the new tested nephelometric method is more precise and gives results that are more reliable because of greater sensitivity and specificity. According to statistical evaluation of linear regression coefficients, Passing Bablock regression line and from the results of parametric statistical tests that we obtained, we can conclude that the present turbidimetric method can be replaced with the nephelometric method and that the methods are comparable.
