Abstract

Za ločevanje lipidov v njihove razrede smo uporabili metodo tankoplastne kromatografije (TLC) in jo po optimizaciji prenesli na metodo ločevanja na trdni fazi (SPE). Vse teste smo izvedli na standardnih spojinah najbolj značilnih predstavnikov lipidov. Rezultate smo ovrednotili na podlagi primerjave kromatogramov, ki smo jih dobili s preizkusi nekaj kombinacij razvijalnih raztopin na TLC in adsorpcijskih faz na SPE. Za našo metodo sta najbolj primerni razvijalni raztopini heksan : dietileter (65 : 35 (v/v)) in kloroform : metanol:voda (65 : 25:4 (v/v/v)), razvijalni raztopini kloroform : metanol:ocetna kislina (100 : 5 : 2 (v/v/v)) in heksan :dietileter : metanol : ocetna kislina (90 : 20 : 5 : 2 (v/v/v)) sta zaradi slabše ločljivosti lipidov manj primerni. Iz rezultatov je razvidno, da je za našo metodo najprimernejša kolona s silikagel polnilom proizvajalca Varian, ki jo kondicioniramo s kloroformom in metanolom. Aminske SPE kolone različnih proizvajalcev niso bile primerne. Uspešni rezultati testov ponovljivosti in obnovljivosti metode so pokazali, da je uporabljena analizna metoda primerna za ločevanje izbranih lipidov v posamezne lipidne razrede.

Secondary title: Separation of lipid class with thin layer chromatography and solid phase extraction
Secondary abstract: Thin layer chromatography (TLC) and solid phase extraction (SPE) were used for separation of lipid classes. After optimization the thin layer chromatographic method was transferred to solid phase extraction (SPE) using standard solutions of lipids. Results were evaluated by comparison of chromatograms obtained with experiments with some possible combination of solutions on TLC and SPE absorption phases. Most suitable developing solutions are hexane:ether (65:35 (v/v)) and chloroform: methanol:water (65:25:4 (v/v/v)), while solutions chloroform:methanol:acetic acid (100:5:2 (v/v/v)) and hexane ether:methanol:acetic acid (90:20:5:2 (v/v/v)) are less appropriate owing of unsuccesful separation. From results it is evident that the most appropriate column is Si – column brand Varian, conditioned with chloroform and methanol. NH2 columns from different manufacturers are not appropriate. Successful results of test repeatability, reproducibility and accuracy of method showed, that applied analytical method is appropriate for lipid separation into individual lipid classes.
