Abstract

Namen diplomske naloge je bil, preučiti vpliv mehanskih sil na izločanje encimov in proteinov iz celic Trimmatostroma salinum pri atmosfereskem tlaku, 35 °C in različnih časih izpostavitve. Rezultati so pokazali, da se je s podaljševanjem časa homogenizacije preživelost celic Trimmatostroma salinum zmanjševala. Določevali smo tudi aktivnost β-glukozidaze, α-amilaze, proteaze, celulaze, ter koncentracijo proteinov pri 595 nm in 280 nm ter merili absorbanco pri 260 nm in 280 nm za določevanje prisotnosti nukleinskih kislin z UV-Vis spektrofotometru. S podaljševanjem časa homogenizacije smo dosegli, da so se iz celic T. salinum izločili intracelularni encimi.

Secondary abstract: The purpose of the diploma thesis was to examine how the secretion of enzymes and proteins are affected by mechanical forces with the extension of homogenization time at 35 °C and atmospheric pressure. The results showed that the survival of Trimmatostroma salinum cells decreased with prolongation of homogenization time at 35 °C. The activity of β-glucosidase, α-amylase, protease, cellulase, concentration of protein at 595 nm and 280 nm, and nucleic acids at 260 nm on UV-Vis spectrophotometer were also measured. With the increase of homogenization time more intracellular enzymes from suspension T. salinum cells were extracted.
