Abstract

Protitelesa spadajo med biološka zdravila, s katerimi se zdravi veliko število bolezni povezanih s poslabšanim imunskim sistemom. Visoka stopnja čistote bistveno prispeva k varnosti in kakovosti teh kompleksnih učinkovin. Afinitetni kromatografski nosilci so v bioprocesništvu ključnega pomena za čiščenje protiteles. Z razvojem novih pristopov za čiščenje tovrstnih molekul, se posledično zmanjšujejo tudi cene bioloških zdravil. Najstarejši afinitetni ligandi so protein A, G in L, ki jih najdemo na površini določenih vrst bakterij. Z napredki na področju rekombinantne tehnologije, proteinskega inženirstva, kombinatorne sintezne kemije in kasneje računalniškega modeliranja organskih spojin, so se razvili afinitetni ligandi z izboljšanimi fizikalno-kemijskimi lastnostmi. Med glavne parametre za industrijsko čiščenje protiteles spada dinamična vezavna kapaciteta afinitetnih kromatografskih nosilcev, na katero vplivamo s povečanjem stabilnosti afinitetnih ligandov, povečanjem afinitete vezave ter mnogimi drugimi dejavniki. Biofarmacevtska podjetja skrbno izberejo proizvajalca afinitetnih kromatografskih kolon, glede na svoj produkt, glede na učinkovitost afinitetnega nosilca ter ceno. Med bolj poznane proizvajalce sodijo Thermo Fischer Scientific, GE Healthcare Life Sciences in Waters Corporation.

Secondary title: Affinity chromatography resins for antibody purification
Secondary abstract: Antibodies are a subclass of biopharmaceuticals used to treat a number of conditions related with impaired immune system. A high degree of purity essentially contributes to safety and quality of these complex substances. Affinity chromatography resins play a vital role in antibody purification process. The development of new approaches in purifying these type of molecules influences on lowering the prices of biopharmaceuticals on the market. The first affinity resins were based on proteins that are found on the surface of certain species of bacteria (protein A, G and L). Due to the progresses in the field of recombinant technology, protein engineering, combinatorial chemistry and later computer modeling of organic compounds new affinity ligands with improved physical and chemical properties were developed. With the improvement of these properties we are able to influence on dynamic binding capacity of the affinity resins, which is one of the main parameters in industrial antibody purification process. Biopharmaceutical companies carefully choose the manufacturer of affinity chromatography columns, regarding to their product, the performance of the affinity resin and its price. Some of the well-known manufacturers are Thermo Fischer Scientific, GE Healthcare Life Sciences and Waters Corporation.
