Abstract

Aloe vera je zdravilna rastlina, ki jo ljudje uporabljajo že tisočletja. Zaradi njenih lastnosti je nepogrešljiva v medicini, kozmetiki, farmaciji in prehrani. V naravi se Aloe vera razmnožuje vegetativno, kar je zelo počasen proces, z nizkim donosom in je mogoč le med rastno sezono. Najprimernejša alternativna oblika razmnoževanja Aloe vere je zato in vitro vegetativno razmnoževanje rastlin v tkivni kulturi, oziroma z mikropropagacijo. Namen naše naloge je bil razviti postopek mikropropagacije Aloe vere, ki vključuje vzpostavitev tkivne kulture, razmnoževanje in koreninjenje poganjkov. Kot začetni izsek smo uporabili mlade stranske poganjke, ki smo jih vcepili na Murashige in Skoog (MS) gojišče, z dodanimi rastnimi regulatorji. Uspešnost mikropropagacije smo merili preko naslednjih parametrov: prisotnost in število brstov, prisotnost in število korenin. Največ poganjkov, v povprečju 5,9, se je tvorilo na MS gojišču z 8 ▫$\mu$▫M BAP in 1 ▫$\mu$▫M NAA, največ korenin, v povprečju 7,2, pa na MS gojišču z 4 ▫$\mu$▫M BAP in 4 ▫$\mu$▫M NAA. Poganjke z dovolj razvitimi koreninami smo prestavili v plastične lončke z zemljo ali mešanico zemlje in peska (1 : 1), kjer se je uspešno aklimatiziralo vsaj 60 % vseh rastlin.

Secondary abstract: Aloe vera is a medicinal plant that people have been using for millenniums. Due to its properties it is indispensable in medicine, cosmetic industry and pharmacy. In nature Aloe vera propagates vegetatively, which is a very slow process, with low income and possible only during growing season. That is why the best suitable method for reproduction of Aloe vera is in vitro vegetative propagation or micro propagation. The purpose of our thesis was to develop a procedure of micro propagation of Aloe vera that includes tissue culture initiation, reproduction and rooting of shoots. For initial shoots we used young lateral shoots that we inoculated in Murashige and Skoog (MS) growth medium, with added growth regulators. The success was measured through the following parameters: the presence and the number of buds, the presence and the number of roots. Most shoots, on average 5.9, were formed on MS medium with 8 ▫$\mu$▫M BAP in 1 ▫$\mu$▫M NAA and most roots, on average 7.2, on MS medium with 4 ▫$\mu$▫M BAP in 4 ▫$\mu$▫M NAA. We moved shoots with sufficiently developed roots to plastic pots with soil or a mixture of soil and sand (1 : 1), where 60 % of all plants were successfully acclimatized.
