Abstract

V tej raziskavi je bil naš namen pregledati in primerjati ultra strukturo celic alfa v Langerhansovih otočkih, v treh makroskopsko razdeljenih delih (spleničnem, gastričnem in duodenalnem delu) trebušne slinavke miši. Opazovali in primerjali smo velikosti glukagonskih veziklov in granul v tkivu, ki je bilo izpostavljeno različnim koncentracijam glukoze v različno dolgih časovnih intervalih. Vzorce tkiva smo analizirali s svetlobno mikroskopijo in presevno elektronsko mikroskopijo (TEM). Dobljene podatke smo statistično ovrednotili. Ugotovili smo, da je ultra struktura organelov celic alfa v različnih delih trebušne slinavke in v tkivu, izpostavljenemu različnim koncentracijam glukoze, primerljiva. Statistično značilne razlike so bile opazne v premerih granul in veziklov ter v površini veziklov med tremi različnimi deli trebušne slinavke. Vzorci, izpostavljeni različnim koncentracijam glukoze v različno dolgih časovnih intervalih, so se statistično razlikovali v premerih granul in veziklov, v površini granul in veziklov ni bilo statističnih razlik. Med premeri in deleži površin granul in veziklov ob plazmalemi, ki so bili izpostavljeni različnim koncentracijam glukoze v različnem času, ni bilo statističnih razlik. Secondary title: Structural characteristic in alpha cells of mouse Langerhans islets
Secondary abstract: In this study, our purpose was to examine and compare the ultrastructure of alpha cells in the islets of Langerhans in three macroscopically divided parts (splenetic, gastric and duodenal region) of the pancreas in mice. We observed and compared the size of glucagon vesicles and granules in the tissue, which was exposed to different glucose concentrations at various time intervals. With light microscopy and transmission electron microscopy (TEM) we analyzed tissue samples. The obtained data were statistically evaluated. We established that the ultrastructure of alpha cells organelles is similar in different parts of the pancreas and at various concentrations of glucose. Statistically significant differences were observed in the diameter of the granules and vesicles, and in the surface of the vesicles between the three different parts of the pancreas. Samples, exposed to different glucose concentrations at various time intervals, statistically differed in the diameter of the granules and vesicles, but no statistical differences were seen in the surface of granules and vesicles. Among the diameters and surface percentage of granules and vesicles near the cell membrane, which were exposed to different glucose concentrations at various time intervals, no statistical differences were seen. For further research, it would be necessary to include a larger number of islets with the contained cell types. Additionally, the treated tissue slices could be exposed to higher concentrations of glucose and for a significant period of time.
