Abstract

Diplomsko delo prikazuje študijo evtektičnih topil in njihovo uporabo za izolacijo hitina iz odpadnih lupin škampov, kvantitativnega in kvalitativnega določanja izoliranega hitina in sintezo hitozana iz pridobljenega hitina za izdelavo biofilmov. Pri študiji smo uporabili štiri različna evtektična topila iz mešanic komponent holin klorid – citronska kislina (CCCA), holin klorid – malonska kislina (CCMA), holin klorid – mlečna kislina (CCLA) in holin klorid – sečnina (CCU) za izvedbo 3-urne in 6-urne fragmentacije 1 g in 2 g zmletih lupin škampov v mešalnem reaktorju pri različnih temperaturah. Za kvalitativno določanje izoliranega hitina smo uporabili FTIR spektroskopijo pri čemer smo dobljen spekter primerjali s spektrom komercialnega vzorca hitina. Morfološke karakteristike izoliranega hitina z uporabo sintetiziranih evtektičnih topil smo nato posneli s SEM mikroskopom. Hitozan smo pridobili preko N-acetilacije z uporabo 70 % raztopine natrijevega hidroksida pri povišani temperaturi. Filme na osnovi pridobljenega hitozana smo naredili z metodo ulivanja v kalup. Najboljše rezultate glede na izkoristek izolacije hitina iz vzorca smo dobili z evtektično zmesjo holin klorid – mlečna kislina (CCLA). V splošnem smo opazili trend povečanja izkoristkov izolacije hitina iz vzorca pri povečanju temperature in mase natehtanih zmletih lupin škampov. Ugotovili smo, da evtektična topila kažejo velik potencial kot okolju neškodljiv in trajnostni nadomestek konvencionalnim topilom, ki so toksična in okolju manj prijazna.

Secondary abstract: The following thesis is a study on eutectic solvents and their usage for isolation of chitin from waste shrimp shells, as well as quantitative and qualitative analysis of the isolated chitin, including the synthesis of chitosan, and its potential for manufacture of biofilms. The study used four different eutectic solvents, including choline chloride-citric acid (CCCA), choline chloride-malonic acid (CCMA), choline chloride – lactic acid, as well as choline chloride – urea (CCU), which were used for 3 hour and 6 hour fragmentation of 1 g and 2 g of ground shrimp shells, at different temperatures. Qualitative analysis was performed with FTIR spectroscopy and X-ray diffraction analysis (XRD). Resulting spectrum of isolated chitin samples was compared with the spectrum of commercial grade chitin. Morphologic analysis of the chitin isolate was then performed with SEM microscopy. Chitosan was synthesised by N-acetylation with the usage of 70% solution of sodium hydroxide at increased temperatures. Chitosan biofilms were then manufactured by casting. Best results, with the best observed efficiency of chitin isolation, were achieved with the eutectic mixture of choline chloride and lactic acid (CCLA). Higher efficiency was also observed at elevated temperatures, and while using higher masses of ground shrimp shells. We find that eutectic solvents show great potential as an environment-friendly, permanent alternative to conventional solvents.
