Abstract

Bakterija Mycoplasma canis pri psih povzroča urogenitalna vnetja in je povezana z neplodnostjo. Mehanizmi patogeneze so slabo raziskani. V okviru magistrske naloge smo z izvedbo gentamicinskega testa določali adhezivne in invazivne sposobnosti bakterije Mycoplasma canis, sev Larissa, na modelu trajne pasje celične linije DH82 in vitro. Rezultati so pokazali sposobnost adhezije Mycoplasma canis na celice DH82 in razkrili tudi invazivne zmožnosti preučevane bakterije, saj v manjšem obsegu mikoplazme vstopajo tudi v gostiteljske celice. Največ Mycoplasma canis se je pritrdilo in vstopilo v celično linijo DH82 po 9 urah kokultivacije, nato pa je število obojih upadalo vse do konca gentamicinskega testa. Mikroskopske slike so pokazale, da okužba privede do morfoloških sprememb na gostiteljskih celicah, ki so najbolj opazne po 48-ih urah inkubacije. Opazimo lahko tudi padec viabilnosti in povečano število liziranih celic DH82, katerih delež je bil največji po 48-ih urah inkubacije. Poleg tega se je kokultuivacija odražala tudi v spreminjanju premera celic DH82, ki se je med poskusom zmanjšal za približno 3 μm. Zmožnost pritrjevanja in invazije v notranjost celic DH82 v in vitro pogojih potrjuje znanstveno domnevo, da se Mycoplasma canis v in vivo sistemih s pripenjanjem in vstopanjem v gostiteljske celice izmika imunskemu odzivu gostitelja, kar vodi v persistentno okužbo, ta pa v pojav kroničnega obolenja, kar pa je značilno za Mycoplasma canis.

Secondary abstract: Canine mycoplasma Mycoplasma canis is associated with urogenital infections and infertility. The mechanisms of pathogenesis are poorly understood. The main goal of this Master's thesis was to determine adhesion and invasion property of studied Mycoplasma canis strain Larissa on the DH82 canine cell line in vitro. Results showed not only the adhesion abilities of Mycoplasma canis, but also, to a lesser extent, the ability to invade into the DH82 host cell line. In both cases, CFU values reached their peak after 9 hours of the incubation period. Microscopic images revealed, that infection of the DH82 cell line with Mycoplasma canis leads to morphological changes, which were most prominent after 48 hours after infection. The slight drop in viability and increase in number of lysed cells is also present and again, most visible after 48 hours of the incubation period. It looks like that infection of DH82 cells with Mycoplasma canis affects the diameter of the infected cells, which is reduced by about 3 μm. Adhesion onto and invasion into the DH82 cells by Mcoplasma canis in vitro confirms scientific presumption, that adhesion and invasion capabilities in vivo are linked with evasion of the hosts immune response by pathogen, leading to persistent infections, and to mild chronic diseases, which are most common among Mycoplasma canis strains.
