Abstract

Perutninsko perje predstavlja enega najbolj obremenilnih in neizkoriščenih odpadkov, saj zavzema do 10 % celotne teže perjadi, kar na letni ravni nanese skoraj 10 milijonov ton odpadkov. Zaradi visokega deleža keratina (90 %) je perje potencialna surovina za pridobivanje le – tega, hkrati pa bi tako postal izkoriščen vir, namesto onesnažujoč odpadek. V sklopu magistrske naloge smo izvedli izolacijo keratina iz perutninskega perja s subkritično vodo, pri temperaturah med 120 °C in 250 °C, in časih med 5 minut in 75 minut. Ugotovili smo, da je optimalna temperatura za izvedbo izolacije 180 °C. S FTIR analizo smo potrdili prisotnost keratina v produktih izolacije s subkritično vodo. Molekulsko maso nastalega produkta smo določili s SDS – PAGE elektroforezo in gelsko kromatografijo. Molekulska masa keratina nastalega pri reakciji izolacije se giblje med 4 kDa in 12 kDa. Iz teh rezultatov sklepamo, da subkritična voda pri visoki temperaturi deluje kot reducent in razcepi disulfidne vezi v molekuli keratina, ki posledično razpade na manjše enote. Za določitev uporabnosti metode izolacije iz perutninskega perja s subkritično vodo v industrijskem merilu, bi bile potrebne nadaljnje raziskave.

Secondary title: Isolation of keratin from feathers using hydrothermal processes
Secondary abstract: Poultry feathers are among the most polluting and unexploited wastes. Feathers represent up to 10 % of chicken weight, which leads to 10 million tons of waste annually. Because of high keratin content (90 %), feathers are a potential raw material for obtaining highly-valuable protein. Therefore, feathers could become a valuable resource, rather than polluting waste. As a part of master’s degree, isolation of keratin from chicken feathers with subcritical water at different temperatures (120 °C – 180 °C) and different times (5 min – 75 min) was applied. Results indicate that 180 °C is optimal temperature for isolation of keratin with subcritical water. Presence of keratin was confirmed using FTIR analysis. Molecular weight of subcritical water isolation product was determined using SDS – PAGE electrophoresis and Size exclusion gel chromatography. The molecular weight of the keratin produced by the isolation reaction ranges between 4 kDa and 12 kDa. In conclusion, subcritical water acts as a reducing agent at higher temperatures, breaking the disulfide bond in the keratin molecule, which consequently decays into smaller units. Further research is needed to determine the viability of the isolation method from poultry feathers with subcritical water on an industrial scale.
