Abstract

Cilj magistrskega dela je bil preučiti kemijsko sestavo skorje bele jelke (Abies alba Mill.) ter določiti antioksidativni in fungicidni potencial pridobljenih ekstraktov. Najprej je bilo potrebno na vsake 14 dni odvzeti skorjo debla. Absolutno suhe vzorce skorje smo zmleli in shranili v prahovke. Ekstrakcija je potekala na ekstraktorju za pospešeno ekstrakcijo. Nato smo naredili kemijsko analizo ekstraktov (gravimetrično analizo, UV-Vis spektrofotometrijo, TLC ter HPLC). Z metodo lovljenja DPPH radikala smo ekstraktom določili antioksidativni potencial. Fungicidni potencial ekstraktov smo določili na 3 testne glive ter 3 plesni. Ugotovili smo, da vlažnost skorje s časom pada. Vsebnost vodotopnih ekstraktivov se ni bistveno spremenila. Zunanji sloj skorje povprečno vsebuje več vodotopnih ekstraktivov, celokupnih fenolov, katehina in epikatehina kot notranji sloj skorje. Višje kot je bil odvzet vzorec skorje na deblu, več vodotopnih ekstraktivov je vseboval. Nižje kot so rastle veje, več vodotopnih ekstraktivov ter celokupnih fenolov je vsebovala njihova skorja. S tankoplastno kromatografijo smo določili, da naš vzorec vsebuje 14 spojin. S pomočjo HPLC analize smo ugotovili, da se v ekstraktih nahaja največ katehina, epikatehina, taksifolina, ferulne kisline in matairezinola. Največji antioksidativni potencial ima ekstrakt pridobljen iz skorje vej. Višja kot je koncentracija ekstrakta, večji je zaviralni učinek rasti gliv in plesni. Ekstrakt, pridobljen iz skorje veje, ima večji inhibitorni učinek na rast gliv in plesni kot ekstrakt, pridobljen iz skorje debla.

Secondary abstract: The goal of the thesis was to study chemical structure of Abies alba Mill's bark and to identify antioxidative and fungicide potential of gathered extracts. At first, we needed to collect samples of trunks bark every two weeks. Absolutely dry samples were ground and placed in reagent bottles. Extraction was executed on accelerated solvent extractor. Then we preformed chemical analysis of extractions (gravimetry analyse, UV-Vis spectrophotometry, TLC and HPLC) and with DPPH method was used to determine the antioxidative potential. Fungicide potential was divided into three mould and three fungus test samples. We discovered that humidity decreases with time and that thebark includes more hydrophilic extractives. On average the outer layer contains more hydrophilic extractives, phenols, catechin and epicatechin than in the inner layer. On average the tree bark samples that were taken higher from the tree, contain more hydrophilic extractives. Branches growing lower on the trunk contain more hydrophilic extractives and phenols in their bark. With TLC we determined that our sample contained 14 compounds. Through HPLC analysis, we found that our extracts contained the most catechin, epicatechin and taxifolin. The extract from the bark of branches has the greatest antioxidative potential. The higher the concentration of the extract, the greater the inhibitory effect on the growth of fungi and mold it has. The extract obtained from the bark of the branches has a greater inhibitory effect than the extract obtained from the bark of the trunk.
