Abstract

Poškodba glave, zastoj srca, hipoksija ali ishemije so možni vzroki, ki vodijo v razvoj citotoksičnega edema centralnega živčnega sistema (patološko otekanje možganskih celic). Astrociti so med prvimi možganskimi celicami, ki otečejo in nadalje povzročijo propad živčnih celic. Zdravila za omejitev njihovega otekanja ter omejitev spremljajočih nevrodegenrativnih posledic ni. Noradrenergična stimulacija, preko aktivacije adrenergičnih receptorjev (α_1-AR in β-AR) na membrani astrocitov, deluje blagodejno, če so ti patološko otekli. Raziskovanje delovanja noradrenalina na celični ravni, je pomemben korak v smeri njegove uporabe kot zdravila za zdravljenje citotoksičnega edema. V magisterski nalogi smo uporabili elektrofiziološko metodo vpete napetosti na krpici membrane v konfiguraciji cele celice. Z njo smo spremljali spremembo kapacitivnosti membrane (C_m) astrocitov iz primarnih celičnih kultur, izoliranih iz možganske skorje 2 do 3 dni starih podgan, pred in po draženju z noradrenalinom. Porast parametra C_m po draženju z noradrenalinom smo pripisali večanju površine plazmaleme, zaradi aktivacije procesa eksocitoze. Znano je, da aktivacija α_1-AR povzroči porast znotrajceličnega kalcija, ki je dražljaj za eksocitozo. Izločanje sekretornih molekul iz veziklov z mehanizmom od kalcija odvisne eksocitoze, so pri astrocitih že zabeležili v predhodnih raziskavah.

Secondary title: Effect of noradrenaline on capacity changing of astrocyte membrane
Secondary abstract: Head injury, stroke, hypoxia or ischemia are all possible causes, which lead to formation of cytotoxic cerebral edema (cell swelling). Astrocytes are among the first brain cell types to undergo swelling, which may lead to neurodegeneration. To this day, there is no medical treatment to reduce astrocyte swelling and formation of neurodegeneration. Noradrenergic stimulation, which activates adrenergic receptors (α-AR in β-AR) on astrocyte plasmalemma, acts favorable to reduce cell edema. Noradrenalin may be a candidate for treatment of cytotoxic edema, however, mechanisms of its action on cell response are not clear yet. We used an electrophysiological method, patch clamp in the whole cell configuration, to better understand processes affecting cell surface fluctuations, after noradrenalin stimulation on cultured cortical rat astrocytes. We have monitored the parameter of cell capacitance (C_m). After stimulation the C_m has increased significantly. It is known that astrocytes secrete their gliotransmitters also in the process of 〖Ca〗^(2+) dependent exocytosis. Noradrenalin activates α_1-AR, which increases intracellular calcium concentration. We concluded that the rise in C_m parameter was due to activation of the process of exocytosis
