Abstract

Bakteriofagi so naravni ubijalci bakterij. Predstavljajo alternativo antibiotikom pri boju z bakterijskimi okužbami. Da bi bila bakteriofagna terapija uspešna, je potrebno poznati bakteriofagni odziv na bakterijo v različnih fizioloških stanjih, ki jih lahko najdemo v človeškem telesu. Bakterije z nizko metabolno aktivnostjo v neugodnem, omejenem okolju povzročajo latentne in trdovratne okužbe. Bakteriofagno inficirane bakterije v okolju z malo hranivi vstopijo v reverzibilno dormantno stanje imenovano hibernacija. Pri eksperimentalnem delu smo s hitrostjo redčenja določali hitrost rasti bakterijske kulture in se v fazi limitacije približevali stacionarni fazi rasti in spremljali bakteriofagno infekcijo in propagacijo v E.coli z nizko metabolno aktivnostjo. Hibernacijski efekt oziroma nenaden porast prostih bakteriofagov je bil dosežen z dodatnim svežih hraniv, s povišanjem bakterijske metabolne aktivnosti.

Secondary title: The influence of physiological state of bacteria Escherichia coli on occurrence of hibernation mode in formation of bacteriophages
Secondary abstract: Bacteriophages are natural killers of bacteria. They represent a plausible alternative to antibiotics. For phage therapy to be successful, we have to learn its effect on bacteria in different physiological states that can be found in human body. Bacteria with low metabolic activity in limiting conditions cause latent and persistent infections. Bacteriophage infected bacteria in limiting conditions enter a reversible dormant state called hibernation mode. In experimental work, dilution rate was used to determine bacterial growth rate. In the phase of limitation we approached the stationary growth phase and studied bacteriophage infection and propagation in E.coli with low metabolic activity. Hibernation effect or sudden increase of free bacteriophages was achieved with the adidion of fresh nutrients correspondingly with the increase of bacterial metabolic activity.
