Abstract

V diplomskem delu je predstavljeno gojenje Pleurotus ostreatus na različnih odpadnih substratih iz kmetijstva z namenom pridobitve visoko aktivnih encimov. Gojišča so bila zasnovana iz rastnega medija, pšeničnih otrobov ter izbranih substratov (slama, trava, bučke,kumare, breskev, marelica, hruška ter paprika). Po osmih dneh inkubacije smo s postopkom ekstrakcije pridobili ekstrakte encimov, ki smo jim določili koncentracijo (vsebnost) proteinov, aktivnost ter specifično aktivnost izbranih encimov celulaze, lakaze ter α- amilaze. Zanimal nas je razrast glive v posameznih vzorcih ter prirast/izguba biomase po osmih dneh gojenja.

Secondary abstract: The diploma presents the cultivation of Pleurotus ostreatus on different waste substrates from agriculture in order to obtain highly active enzymes. The media were designed from growing medium, wheat bran and selected substrates (straw, grass, courgettes, cucumbers, peaches, apricots, pears and peppers). After eight days of incubation, enzyme extracts were obtained by the extraction process, which determined the concentration (content) of proteins, activity and specific activity of selected enzymes (cellulase, laccase and α-amylase). We were interested in the growth of the fungus in individual samples and we observed the increase / loss of biomass after eight days of cultivation.
