Abstract

V magistrskem delu smo raziskovali možnosti za hrambo ozkolistnega koščca (Berula erecta) v tkivni kulturi z upočasnjeno rastjo. Upočasnitev rasti smo poskušali doseči z nižanjem temperature in dodajanjem sladkorjev in sladkornih alkoholov na gojišče MS. Ugotavljali smo vpliv nizkih temperatur (13 °C in 4 °C), različnih kombinacij in koncentracij sorbitola (20, 40 g l-1) in saharoze (30, 40 g l-1) ter svetlobe na viabilnost, rast in barvila ozkolistnega koščca. Spremljali smo različne rastne parametre, vsebnost fotosinteznih barvil in antocianinov. Povišanje koncentracije saharoze ni zavrlo oziroma upočasnilo rasti. Pri 23 % 2 °C je dodatek 20 g l-1 sorbitola uspešno zavrl rast, dodatek 40 g l-1 sorbitola pa je po dveh mesecih kulture vodil v senescenco rastlin. Hramba Berule erecta pri 13 °C v nobenem tretmaju ni bila učinkovita. Kot najbolj učinkovit način za dolgoročno hrambo B. erecta z minimalno rastjo in visoko viabilnostjo se je izkazala hramba rastlin pri 4 °C v temi.

Secondary abstract: In the master's thesis we investigated the possibilities for slow growth storage of Berula erecta. By lowering the temperature and adding sugars and sugar alcohols to the MS medium we attempted to reach growth retardation. We researched the influence of low temperatures (13 ° C and 4 ° C), various combinations of both, concentrations of sorbitol (20, 40 g L-1) and sucrose (30, 40 g L-1) and light on the viability, growth and photosynthetic pigments of B. erecta. We monitored various growth parameters, photosynthetic pigments and anthocyanins. The increase of sucrose concentration did not inhibit or decelerate growth. At 23 % 2 ° C, the addition of 20 g L-1 sorbitol successfully inhibited growth, and the addition of 40 g L-1 sorbitol led to plant senescence after two months of in vitro storage. Storage of Berula erecta at 13 ° C was not effective in any treatment. Storing the plants at 4 ° C in the darkness proved to be the most effective long-term way to store B. erecta with minimal growth and high viability.
