Abstract

Ledeniki so dolgo veljali za abiotska okolja. Novejše raziskave pa kažejo, da so ledeniki posebni in dinamični mikrobni habitati, kjer uspevajo različni psihrofilni in psihotolerantni mikroorganizmi. Zaradi globalnega segrevanja se visokogorski ledeniki pospešeno talijo. S tem izginjajo okolja, v katerih so se več tisočletij uspešno razvijale mikrobne združbe v relativno stabilnih pogojih. Mikroorganizmi, ki naseljujejo ledenike, so razvili vrsto prilagoditev na življenje pri nizkih temperaturah in na ciklična zamrzovanja in odtaljevanja. V Sloveniji trenutno obstajata dva ledenika. Triglavski ledenik leži v Julijskih Alpah, na severovzhodnem pobočju najvišjega vrha v državi, Triglava. Ledenik pod Skuto leži dobrih petdeset kilometrov vzhodneje ter nekaj 100 m južneje od Triglavskega ledenika. Vzorčenje na obeh ledenikih je potekalo v septembru 2018. Iz obeh ledenikov smo aseptično odvzeli tri vzorce ledu iz treh različnih lokacij, jih stalili in filtrirali ter filtre inkubirali na štirih različnih gojiščih primernih za rast gliv in na enem gojišču za bakterije pri temperaturi 15 °C. Skupno smo osamili 181 glivnih izolatov, ki se uvrščajo v 19 rodov, in 66 bakterijskih izolatov, ki se uvrščajo v 22 rodov.

Secondary abstract: Glaciers were long known as abiotic environments, but latest reseach shows that they are special and dynamic habitats where various psychrofilic and psychrotolerant organisms can thrive. Due to global warming highland glaciers are rapidly thawing, releasing enclosed cold-adapted microbial communities into the environment. Since glacial microbial communities were growing in relatively stable conditions, characterized by low temperature, few nutrients, surface exposure to UV-radiation, increased salinity in brine channels around ice crystals, many can't survive these changes. In Slovenia, only two glaciers (glacierets) remain. Triglav glacier is located in Julian Alps, on the northeast slope of the country's highest peak, Mt. Triglav. Skuta glacier, a bit further to the east is located in Kamnik-Savinja Alps. Sampling of ice was conducted on both glaciers in September 2018. Three samples on different locations on each glacier were taken and brought to the laboratory for further analyses. The samples were aseptically thawed and filtrated through membrane filters and incubated on different selective microbial culture media with temperature of 15 °C. We succsefully isolated and identificated 181 strains of fungi belonging to 19 genera and 66 bacterial strains belonging to 22 genera.
