Abstract

Magistrska naloga je zajemala preučevanje inhibitornih lastnosti ekstraktov zmletih koščic asimine (Asimina triloba) ter granatnega jabolka (Punica granatum L) na rast različnih mikrobnih kultur. Pripravili smo različne ekstrakte ter preizkušali njihov inhibitorni vpliv na rast različnih mikroorganizmov kot so: Escherichia coli, Pseudomonas fluorescens, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Streptococcus pyogenes, Saccharomyces cerevisiae, Streptomyces platensis, Candida albicans, Aspergillus flavus, Aspergillus brasiliensis, Aspergillus niger, Aspergillus fumigatus, Penicillium cyclopium in Trichoderma viride. V ekstraktih zmletih koščic asimine smo določili tudi vsebnost totalnih fenolov, proantocianidinov ter njihovo antioksidativno aktivnost. Inhibitorno učinkovitost vzorcev smo proučevali z dvema metodama: z disk difuzijsko metodo ter dilucijsko metodo v bujonu. Z disk difuzijsko metodo smo proučili kateri vzorci izkazujejo inhibitorne lastnosti. Vzorce, ki so izkazali inhibitorne lastnosti, smo nato analizirali še z dilucijsko metodo v bujonu. Za to metodo smo uporabili sledeče mikrobne kulture: E. coli, P. fluorescens, P. aeruginosa, S. aureus, B. cereus, S. pyogenes in C. albicans. Tako smo določili minimalno inhibitorno koncentracijo ekstrakta ter stopnjo inhibicije rasti mikrobnih kultur po določeni inkubacijski dobi. Rezultati raziskovalnega dela so pokazali, da ekstrakti zmletih koščic asimine ne izkazujejo dobrih inhibitornih lastnosti. Z dilucijsko metodo smo ugotovili, da vsi testirani ekstrakti granatnega jabolka delujejo inhibitorno na rast vseh testiranih mikroorganizmov. Kot najboljši inhibitor rasti izbranih mikrobnih kultur se je izkazal liofiliziran vodni ekstrakt liofiliziranih lupin granatnega jabolka, ki je uspešno zaviral rast 5 različnih mikrobnih kultur (B. cereus, E. coli, P. fluorescens, P. aeruginosa in S. pyogenes). Najnižjo stopnjo inhibicije rasti pa smo določili za C. albicans.

Secondary abstract: The purpose of the master's thesis was to study the inhibitory properties of extracts of asimina (Asimina triloba) and pomegranate (Punica granatum L.) on the growth of various microbial cultures. We prepared different extracts and tested their inhibitory properties on different microbial cultures such as: Escherichia coli, Pseudomonas fluorescens, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Streptococcus pyogenes, Saccharomyces cerevisiae, Streptomycus, Streptomyces niger, Aspergillus fumigatus, Penicillium cyclopium and Trichoderma viride. Asimine extract was used to determine the content of total phenols, proanthocyanidins and antioxidant activity. Antimicrobial efficacy of the samples was studied by two methods: the disk diffusion method and the dilution method. First, we used a disk diffusion method to check which samples exhibit inhibitory properties. Samples exhibiting inhibitory properties were then analyzed by the dilution method. Analyzes were performed on the following microbial cultures: Escherichia coli, Pseudomonas fluorescens, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Streptococcus pyogenes, and Candida albicans. This method was used to determine the minimum inhibitory concentration and the degree of growth inhibition of microbial cultures after a certain incubation period. We found that extracts of ground asimine seeds do not exhibit good inhibitory properties. Using the dilution method, we found that all tested pomegranate extracts had an inhibitory effect on the growth of all tested microorganisms. The best inhibitor was lyophilized water extract of lyophilized pomegranate peels, which proved to be the best inhibitor in 5 different microbial cultures (B. cereus, E. coli, P. fluorescens, P. aeruginosa and S. pyogenes). The lowest degree of inhibition was determined by the fungus C. albicans.
