Abstract

Konoplja je v zahodnem svetu dobila pomembnejši pomen po letu 1851, ko je bila vključena v takratno ameriško farmakopejo. V zadnjem času se na trgu pojavlja vedno več prehranskih in kozmetičnih izdelkov, ki vsebujejo kanabinoide. Predvsem je govora o terapevtskih in protibolečinskih učinkih določenih kanabinoidov, na primer CBD, CBG, CBN. Glede na evropske smernice, vsebnost psihoaktivnega ∆^9-THC v končnih izdelkih ne sme presegati 0,2 % v suhi snovi rastline. Za zagotavljanje varnosti in učinkovitosti pripravkov iz konoplje je potrebna ustrezna karakterizacija izdelkov in kvantifikacija učinkovin v njih, zlasti določitev zakonsko spornega ∆^9-THC. V magistskem delu smo se ukvarjali z razvojem metod za določitev sledi ∆^9-THC z različnimi kromatografskimi metodami z namenom nadzora kvalitete in stabilnosti izdelkov iz naravnih in sintetičnih kanabinoidov. Razvili in validirali smo kromatografsko metodo za določitev sledi ∆^9-THC s HPLC-UV/VIS. Z namenom razvoja bolj občutljive metode smo nato na sistemu LC-MS/MS optimizirali parametre masnega spektrometra in razvili ter delno validirali metodo. Z omenjenima metodama so bili rezultati določitve ∆^9-THC v vzorcu CBD primerljivi. Preizkusili smo tudi uporabo GC FID, vendar se ta za določitev zelo nizkih koncentracij ∆^9-THC ni izkazal kot ustrezen.

Secondary abstract: In the Western world, cannabis gained more importance after 1851, when it was included in the American Pharmacopoeia. Recently, more and more food and cosmetic products containing cannabinoids are appearing on the market. In particular, there is talk of therapeutic and analgesic effects of certain cannabinoids, such as CBD, CBG, CBN. According to European guidelines, the content of psychoactive ∆^9-THC in the final products should not exceed 0,2 % in the dry matter of the plant. To ensure the safety and efficacy of cannabis preparations, it is necessary to properly characterize the products and quantify the active substances in them, in particular to determine the legally controversial ∆^9-THC. In the master's thesis, we developed methods for determination of ∆^9-THC in traces by various chromatographic methods for quality and stability control of natural and synthetic cannabinoid products. Using HPLC-UV/VIS, a chromatographic method to determine traces of ∆^9-THC was developed and validated. In order to develop more sensitive method, we optimized the parameters of the mass spectrometer on the LC MS/MS system and developed and partially validated the method. The results of the determination of ∆^9 THC in CBD sample by the mentioned methods were comparable. The use of GC FID was also tested, but it was not suitable for determining such low ∆^9 THC levels.
