Abstract

V nalogi smo z metodo slikovne citometrije analizirali velikost jedrnega genoma kačnikovk, ki smo jih nabrali v La Gambi (Kostarika). Ugotavljali smo, ali imajo rastline z isto življenjsko obliko podobne velikosti genomov, ali imajo sorodne vrste podobne velikosti genomov in ali velikost genoma kaže, da je v raziskovanih vzorcih prisotna poliploidija. Izmerili smo velikost genoma 46 vrst iz 12 rodov, pri čemer smo ugotovili velik razpon vrednosti 2C. Pridobili smo prve podatke za 39 vrst in 3 rodove. Potrdili smo, da življenjska oblika raziskovanih vrst ni povezana z velikostjo genoma, obstaja pa povezava med velikostjo genoma in življenjsko dobo. Vrste v ožjem sorodstvu so imele podobne velikosti genomov le pri rodovih Philodendron in Pistia. Poliploidijo smo ovrgli pri taksonu Philodendron sp. (vzorec Etl #9), za vrsto Philodendron cf. sulcatum pa bi potrebovali dodatne raziskave. Prešteli smo število kromosomov nedoločene vrsterodu Adelonema (2n = 10), kar je prvi podatek za ta rod. Za 5 vrst priporočamo dodatne meritve zaradi metodoloških pomankljivosti, ostale meritve pa so primerne za vnos v spletno bazo vrednosti C (Royal Botanic Gardens, Kew, VB). Naše meritve smo primerjali z znanimi podatki kačnikovk, enokaličnic in kritosemenk na splošno ter ugotovili, da imajo kritosemenke v povprečju manjši genom kot kačnikovke, čeprav so taksonomska nadkategorija, ki vključuje kačnikovke in enokaličnice.

Secondary title: Determining the amount of nuclear DNA of selected tropical species in the Arum family (Araceae)
Secondary abstract: We measured nuclear genome size of aroids, collected in La Gamba (Costa Rica), using DNA image cytometry. We aimed to establish whether plants with the same life form have similar genome size, whether closely related species have similar genome sizes and whether polyploidy is present in any of the studied samples. We measured 46 species from 12 genera, in which we found a wide range in 2C-values. 39 species and 3 genera were measured for the first time. We determined that life form is not correlated with genome size, while life cycle is. Closely related species had similar genome sizes only in genera Philodendron and Pistia. Polyploidy was disproved for taxon Philodendron sp. (sample Etl #9), while additional research is necessary for Philodendron cf. sulcatum. We also counted the chromosome number for an unidentified species of genus Adelonema (2n = 10), which is the first data for this genus. Due to methodological flaws, we recommend additional measurements for 5 species, while other measurements could be included into the online C- value database by Royal Botanic Gardens, Kew (Great Britain). Our measurements were compared with known data of aroids, monocots, and angiosperms in general. We found angiosperms have on average a smaller genome size than aroids, despite being the clade that includes both aroids and monocots.
