Abstract

Elektroporacija je fizikalna metoda, ki omogoča prehajanje številnih molekul skozi celično membrano, ki jih celica v normalnih pogojih ne prepušča. Z uporabo kratkih, visokonapetostnih električnih pulzov v plazemski membrani povzročimo nastanek hidrofilnih por. Ločimo reverzibilno elektroporacijo, po kateri se celična membrana zaceli in ireverzibilno elektroporacijo, po kateri se celična membrana ne more regenerirati ter posledično pride do celične smrti. Elektroporacija ima veliko možnih aplikacij na več področjih. Uporablja se jo za zlivanje celic, transfekcijo ali transformacijo celic, sterilizacijo vode, pasterizacijo hrane, ekstrakcijo snovi. Elektroporacija je dobila tudi mesto v medicini, na področju zdravljenja raka. Za ta namen lahko uporabimo reverzibilno elektroporacijo, s katero omogočimo, da v rakaste celice prodrejo citostatiki in tako povečamo njihov učinek. Pri uporabi citostatika cisplatin z elektroporacijo povečamo njegovo učinkovitost za 70-krat, pri bleomicinu pa kar do 700-krat. Možna je tudi uporaba ireverzibilne elektroporacije, kjer rakaste celice ubijemo in jo uporabljamo za netermično ablacijo tkiv in tumorjev.

Secondary title: Use of electroporation in the destruction of cancer cells
Secondary abstract: Electroporation is a physical method that allows the passage of many molecules through the cell membrane that is impermeable to this very molecules. Using short, high-voltage electrical pulses in the plasma membrane causes the formation of hydrophilic pores. Membrane electroporation can either be reversible (cell membrane reseal) or irreversible (cell membrane cannot return to its primary state and cell death occurs). Electroporation has many possible applications in several areas. It is used for cell fusion, water sterilization, food pasteurization, substance extraction, transfection, transformation, etc. Electroporation has also gained a place in medicine, especially in the field of cancer treatment. For this purpose, reversible electroporation is used to introduce cytostatic into cancer cells. When electroporation is used, the efficiency of cisplatin increases 70 times and the efficiency of bleomycin up to 700 times. We can also use irreversible electroporation, where we kill cancer cells, for non-thermal ablation of tissues and tumors.
