Abstract
Rastlinske bioaktivne spojine (antioksidanti, totalni fenoli, proantocianidini, flavonoidi, itd.) so sekundarni metaboliti, ki jih proizvajajo rastline in izzovejo farmakološke ali toksične učinke pri človeku in živalih. Postopek priprave in izbira topila vplivata na kvaliteto in kvantiteto ekstrakta oz. komponent v ekstraktu.
V magistrski nalogi smo raziskali rastlino Arnico Montano, ki ima v farmacevtski in kozmetični industriji velik potencial. Osredotočili smo se na postopek superkritične ekstrakcije in določili optimalne pogoje superkritične ekstrakcije arnike s CO2 na podagi izrisanega eksperimentalnega načrta, programa Design Expert Pro 12. Ugotovili smo, da višja kot sta temperatura in tlak, višji je izkoristek ekstrakcije. Pridobljenim ekstraktom smo s spektrofotometrično metodo determinirali biološko aktivne komponente (antioksidanti, totalni fenoli, proantocianidini, totalni flavonoidi), katerih prisotnost naj bi vplivala na protivnetni, antiradikalni, antibakterijski, protiglivični in antioksidativni potencial. Rezultati so nam pokazali, da ekstrakti, ki so bili pridobljeni pri višji temperaturi ter tlaku, vsebujejo višje vrednosti biološko aktivnih komponent. Slednjim smo nato določali še protimikrobni potencial, ki smo ga potrdili le ekstraktu 8 z najvišjo vsebnostjo antioksidantov ter totalnih flavonoidov, pridobljenem pri tlaku 100 bar in temperaturi 60 C.

In our master's thesis, we researched the Arnico Montano plant, which has great potential in the pharmaceutical and cosmetic industries. We focused on the supercritical extraction process and determined the optimal conditions for supercritical extraction of arnica with CO2 on the basis of a plotted experimental plan, Design Expert Pro 12. We found that the higher the temperature and pressure, the higher the extraction efficiency. The obtained extracts were determined by the spectrophotometric method of biologically active components (antioxidants, total phenols, proanthocyanidins, total flavonoids), the presence of which is expected to affect the anti-inflammatory, antiradical, antibacterial, antifungal and antioxidant potential. The results showed us that the extracts obtained at higher temperature and pressure contain higher values of biologically active components. The latter were then determined for their antimicrobial potential, which was confirmed only in extract 8 with the highest content of antioxidants and total flavonoids, obtained at a pressure of 100 bar and a temperature of 60 C. |
