Abstract

Sintetiziranih, izoliranih in karakteriziranih je bilo šest novih bifenilnih spojin na osnovi analoga [18F]FDDNP z vsebovanim push-pull sistemom. Njihovi raznoliki optični spektri, pripadajoči kvantni izkoristki in absorpcijski koeficienti izkazujejo izjemen vpliv elektron-privlačnih skupin na optične lastnosti molekulskih sond. Ugoden batokromni premik absorpcijskega in emisijskega maksimuma sintetiziranih molekul je zaželjen pri detekciji proteinskih agregatov, povezanih z Alzheimerjevo boleznijo, s fluorescenčno mikroskopijo, saj omogoča manjšo lastno fluorescenco. Poleg tega njihov velik Stokesov premik povečuje razmerje signal/šum med detekcijo. Ugodne optične lastnosti molekul skupaj z njihovo paličasto obliko nakazujejo njihovo potencialno uspešno uporabo v diagnostiki Alzheimerjeve bolezni.

Secondary title: Synthesis of biphenyl-based derivatives with interesting solvatochromic properties
Secondary abstract: In this work we successfully synthesized, isolated and characterized six new biphenyl compounds based on the [18F]FDDNP analogue, containing a push-pull system. Their diverse optical spectra, quantum yields, and molar absorptivity demonstrate the extraordinary influence of electron-withdrawing groups on the optical properties of molecular probes. Synthesized probes exhibit a favorable bathochromic shift of the absorption and emission maxima, allowing for less autofluorescence in the detection of protein aggregates connected to Alzheimer’s disease using the fluorescence microscopy analysis. Furthermore, their large Stokes shifts increases the signal-to-noise ratio during the detection. The favorable optical properties of the synthesized molecules, together with their rod-like shape, suggest their potentially successful use in the diagnosis of Alzheimer’s disease.
