Izolacija in karakterizacija bakterij iz rizoplana hladnikije

Abstract

Na površini korenin, imenovani rizoplan, lahko najdemo zelo različne skupine mikroorganizmov. Ti lahko na rast rastlin vplivajo pozitivno, lahko pa s svojo prisotnostjo in delovanjem rastlini škodujejo. Cilj diplomskega dela je izolacija in identifikacija bakterij, ki naseljujejo rizoplan hladnikije - slovenske endemične vrste. Iz treh rastlin smo osamili 17 bakterijskih sevov. Izolirali smo bakterijsko DNA in s pomočjo verižne reakcije s polimerazo (PCR) pomnožili 16S rDNA. Glede na primerjavo pomnožene 16S rDNa z referenčnimi bazami podatkov (GenBank, Ribosomal database project) smo izolirane bakterije identificirali. Izolirane bakterije so pripadale rodovom Bacillus, Enterobacter, Pseudomonas, Erwinia in Pantoea.

Secondary abstract:	On the surface of roots called rhizoplane various groups of microorganisms are present. They can have plant growth promoting effects, but their presence and activity can also have inhibit the plant growth. The aim of the thesis was the isolation and identification of bacteria, which dwell in the rhizoplane of the Slovene endemic genus Hladnikia. From three plants we isolated17 bacterial strains. We isolated the DNA from the bacteria and amplified 16S rDNA regions with polymerase chain reaction (PCR); The identification of the strains was done by compariosn of 16S rDNa sequences with reference databases GenBank and Ribosomal Database Project. The isolated bacteria belonged to the genera Bacillus, Enterobacter, Pseudomonas, Erwinia and Pantoea.
