Abstract

Raziskali smo vpliv sterilizacije, mineralne sestave in strjevanje gojišča na kalitev spor in zgodnji razvoj gametofitov praproti Platycerium bifurcatum. Preučili smo, kako na kalitev spor praproti P. bifurcatum vplivajo razlike v mineralni sestavi Knopovega, dveh različkov Knudsonovega, Moorovega, polovičnega Murashige-Skoogovega in Thompsonovega gojišča, ki jih raziskovalci uporabljajo za kalitev spor drugih vrst praproti, ter jih primerjali s kalitvijo spor na Knopovem in Murashige-Skoogovem gojišču, ki sta najpogosteje omenjeni gojišči za kalitev spor P. bifurcatum. Način sterilizacije spor vpliva na kalitev spor. Spore so kalile na vseh uporabljenih gojiščih. Kaljivost spor je bila podobna na vseh gojiščih različne mineralne sestave, ne glede na prisotnost strjevanja gojišča. Mineralna sestava gojišča in prisotnost strjevanja gojišča vplivata na velikost celic in dolžino rizoidov v zgodnjem razvoju gametofita. Na gojiščih z dodanim strjevanjem gojišča se je razvilo manjše število daljših rizoidov in večje število manjših celic. V tekočih gojiščih se je razvilo večje število krajših rizoidov in manjše število večjih celic.

Secondary abstract: We studied the influence of sterilization, mineral composition and medium hardener on germination and early development of the gametophytes of the fern Platycerium bifurcatum. We have considered how the germination of spores of the fern P. bifurcatum is influenced by differences in mineral composition of the Knop, two varieties of Knudson, Moore, half Murashige-Skoog and Thompson medium, the most frequently used media for germination of other species of fern spores, and compared them with the germination of the spores on the Knop and Murashige-Skoog medium, the most frequently mentioned mediums for germination of P. bifurcatum. The maner of sterilization affects spore germination. Spores germinated on all used media. Germination of the spores was similar on all media with different mineral composition, with or without media hardener. Mineral composition of the media and the presence of medium hardener affect the size of cells and length of rhizoids in early development of gametophytes. On media with added medium hardener, gametophytes developed smaller number of longer rhizoids and a larger number of smaller cells. In liquid media they developed higher number of shorter rhizoids and smaller number of larger cells.
