Abstract

Raziskali smo vpliv jasmonske kisline (JA) in ranitve ter sočasni učinek obeh dejavnikov na regeneracijo listov praproti Platycerium bifurcatum v tkivni kulturi. V MS gojišče z 0,01 % saharoze ter z 0, 0.1, 1, 10 in 100 % M JA smo polagali neranjene ločene liste in ločene liste, ki smo jih različno močno ranili. Po 50 dneh kulture smo na listih spremljali razvoj novih struktur: brstov, rizoidov, apospornih gametofitov in intermediatov. Ugotovili smo, da nižje koncentracije JA pospešujejo razvoj brstov, rizoidov ter intermediatov, medtem ko višje koncentracije vzpodbudijo razvoj apospornih gametofitov. Na listih z ranjeno zgornjo povrhnjico se je razvilo največje število sporofitskih struktur (brstov in rizoidov) in intermediatov, medtem ko je bil razvoj apospornih gametofitov najbolj pospešen na nasekljanih listih. Regeneracija listne kulture pod vplivom različnih koncentracij eksogeno dodane JA je bila podobna regeneraciji, sproženi kot odziv na različne stopnje ranitve.

Secondary abstract: The effect of jasmonic acid (JA), wounding and simultaneous influence of both parameters on regeneration of Platycerium bifurcatum fern leaves in tissue culture were studied. Intact separate leaves and separate leaves that were wounded to varying degrees were placed flat on MS medium supplemented with 0.01 % sucrose and with 0, 0.1, 1, 10 in 100 % M JA. After 50 days of culture new structures developed on leaves: buds, rhizoids, aposporous game-tophytes and intermediates. We found out that low JA concentrations promote development of buds, rhizoids and intermediates, while higher concentrations stimulate development of aposporous gametophytes. The largest number of sporofitic structures (buds and rhizoids) and intermediates developed on leaves with cutted adaxial epidermis, while development of aposporus gametophytes was largely accelerated on chopped leaves. Regeneration of leaf culture affected by exogenously added different concentrations of JA was similar to regene-ration, initiated in response to different levels of wounding.
