Abstract
Namen diplomskega dela je bila priprava kompozitov, sestavljenih iz hitozanskih aerogelov in pen polikaprolaktona.
V sklopu eksperimentalnega dela smo najprej pripravili hitozanske aerogele v obliki kroglic. Gele smo tvorili s sol-gel sintezo, in jih nato posušili s superkritičnim ogljikovim dioksidom. Dobljene aerogele smo uporabili za pripravo kompozitov, ki smo jih formirali s tehniko superkritičnega penjenja. Penjenje smo izvedli pri dveh različnih pogojih, in sicer pri 150 bar/40 °C ter pri 200 bar/60 °C. Pri vsakem pogoju smo pripravili tri kompozite z različnimi deleži aerogelov (2,5 ut. %, 5 ut. %, 10 ut. %) in eno čisto peno. Pripravljene kompozite in peni smo karakterizirali s plinsko adsorpcijo, termično analizo (TGA/DSC), optičnim mikroskopom, vrstičnim elektronskim mikroskopom in Fourierevo transformirano infrardečo spektroskopijo ter določili njihove deleže nabrekanja v simulirani telesni tekočini. Na koncu smo izbrali kompozit z najboljšimi lastnostmi in ga superkritično impregnirali z indometacinom.
Rezultati kažejo, da se lastnosti kompozitov v primerjavi s penami izboljšajo. V nadaljevanju pogoji, pri katerih smo pripravili kompozite, so značilno vplivali na njihove končne lastnosti. Nenazadnje, impregnacija kompozitov z aktivno učinkovino je bila uspešna

The aim of the thesis was to prepare composite materials from chitosan aerogel and polycaprolactone foams.
Firstly, chitosan aerogel beads were prepared. Gels were formed following sol-gel synthesis and drying with supercritical CO2. Prepared aerogels were further used to make composites using the supercritical foaming technique. Foaming was performed using different conditions: 150 bar/40 °C and 200 bar/60 °C. Three different composites were prepared with varying contents of aerogel (2,5 wt%; 5 wt%; 10 wt%) and one pure foam for each set of conditions. Composites and foams were then characterized with N2 gas adsorption, thermal analysis (TGA/DSC), stereo microscope, scanning electron microscope, and FTIR spectroscopy. Furthermore, the swelling ratios were determined in a simulated body fluid. Lastly, the composite with the best properties was supercritically impregnated with indomethacin.
Results showed superior properties of composites compared to the foams. Furthermore, the impregnation with an active pharmaceutical ingredient was also successful. |
