Abstract

Encimi imajo v industriji pomembno vlogo kot okolju prijazni biokatalizatorji. Z imobilizacijo lahko podaljšamo čas njihove uporabe v procesih, izboljšamo stabilnost pri ostrih industrijskih pogojih in izvajamo kontinuirne procese. Ker pa je potrebna tudi dovolj visoka čistost encimov, jih je pred tem potrebno izolirati in očistiti, kar pa predstavlja zahteven in drag korak. Temu se lahko izognemo z enostopenjsko imobilizacijo encimov. Pogosta tehnika direktne imobilizacije encimov je vezava preko označevalcev. Takšne tehnike so visoko specifične, saj vez z nosilcem tvorijo le označeni encimi. Aktivnost imobiliziranih encimov je sicer nižja od aktivnosti prostih, vendar pa imobilizirane biokatalizatorje lahko uporabimo večkrat in jih lažje ločimo od reakcijske mešanice. V diplomski nalogi je predstavljen pregled literature na področju enostopenjskega čiščenja in imobilizacije encimov.

Secondary title: One-step purification and immobilization of enzymes
Secondary abstract: Enzymes play a crucial role in industry as environmentally friendly biocatalysts. Immobilization can extend the time they can be used in processes, improve stability under harsh industrial conditions and enable continuous use. Since enzymes also need to be sufficiently pure, they need to be isolated and purified, before immobilization, which is a challenging and costly step. This can be avoided by one-step immobilization of enzymes. A common technique of direct enzyme immobilization is via tags. Such technique are highly specific, as only the tagged enzymes bond on the carrier. Although the activity of immobilized enzymes is lower than that of free enzymes, immobilized biocatalysts can be used repeatedly and can be easily separated from the reaction mixture. In this thesis a literature review of one-step purification and immobilization of enzymes is presented.
