Abstract

Namen naloge je uporabiti molekularne tehnike za potrditev morfološke diferenciacije že opisanih podvrst L. hochenwartii. Z vzorci smo pokrili celoten areal vrste. Pridobili smo zaporedja dveh odsekov CO1 in izračunali filogenetski drevesi z metodo največjega verjetja in z Bayesovim pristopom. Z metodama ASAP in mPTP, smo izračunali število molekularnih operacijskih taksonomskih enot (MOTU), rezultate mPTP pa smo dodatno preverili z izračunom povprečnih p- razdalj med MOTUji. Podobnost med skupinami smo ugotavljali tudi z mrežo haplotipov. Tako filogenetska drevesa, kot razmejitvene metode ločijo vzorce na skupine, ki po geografski razširjenosti sovpadajo s trenutno opisanimi podvrstami. Z rezultati smo tako potrdili obstoj nekaterih podvrst, glede na izračunane povprečne p-razdalje bi te podvrste lahko bile tudi samostojne vrste. Našli smo dve novi skupini, ki verjetno pripradata še neopisanim podvrstam. Rezultati raziskave, bodo v prihodnosti lahko služili kot osnova za ločitev podvrst na vrste, ko tudi pri nadaljnih odločitvah glede varovanja taksona.

Secondary title: Intraspecific genetic variability of the slender-necked beetle in the Dinaric karst
Secondary abstract: The aim of our research was to use molecular techniques to confirm the morphological differentiation of already described subspecies of L. hochenwartii. Samples covered the entire area of the species. Sequences of two sections of CO1 were obtained, and phylogenetic trees were calculated, using the maximum likelihood method and the Bayesian approach. Using the ASAP and mPTP methods, the number of molecular operational taxonomic units (MOTUs) were calculated, and mPTP results were further verified by calculating the average p- distances between MOTUs. Similarity between groups was also determined using the haplotype network. Both phylogenetic trees and delimitation methods separate samples into groups whose geographical distribution coincides with the currently described subspecies. The results confirmed the existence of some subspecies, according to the calculated average p-distances, these subspecies could also be independent species. We found two new groups that probably belong to so far undescribed subspecies. The results of the research will be able to serve as a basis for the separation of subspecies into species, as well as for further decisions regarding the protection of the taxon.
