Abstract

V diplomskem seminarju z matematičnim modelom presnove arahidonske kisline preučujemo vpliv nesteroidnih antirevmatikov (NSAR) na pojav bronhokonstrikcije pri aspirinsko intolerantnih astmatikih. Za tri različne populacije aspirinsko-intolerantnih astmatikov ocenimo, pri kateri dozi indometacina in etoricoxiba se tveganje za simptome aspirinske intolerance poveča in ocenimo, kako dolgo traja bronhokonstrikcija pri tipični terapevtski dozi indometacina. Preučujemo tudi strategijo, pri kateri aspirin, ibuprofen ali indometacin doziramo skupaj z inhibitorjem encima 5-lipoksigenaze. Ta strategija bi lahko omogočila varno doziranje nesteroidnih antirevmatikov aspirinsko-intolerantnim astmatikom brez tveganja bronhokonstrikcije.

Secondary abstract: In this diploma work we studied the impact of non-steroidal anti-inflammatory drugs (NSAIDs) on the occurrence of bronchoconstriction in aspirin-intolerant asthmatic patients. For this purpose a mathematical model is used to describe the interaction between NSAID with arachidonic acid metabolism in white blood cells. We estimated, for three different populations of aspirin-intolerant asthmatic patients, at which doses of indomethacin and etoricoxib the risk of bronchoconstriction is increased. In addition, the duration of bronchoconstriction is theoretically estimated for typical therapeutic dose of indomethacin. We also study the strategy for safe managing of NSAIDs to aspirin-intolerant asthmatic patients in combination with inhibitor of the enzyme 5- lipoksigenaze (5-LOX) without risk of bronchoconstriction.
