Abstract

Raziskali smo začetek aktivnosti v kambijevi coni navadne bukve (Fagus sylvatica L.) na 2 rastiščih Panška reka (400 m n.m.v.) in Menina planina (1200 m n.m.v). Na 6 drevesih z vsakega rastišča smo spremljali začetek diferenciacije celic in nastajanje ranega lesa ksilemske branike 2008. V tedenskih intervalih, od 17. marca do 23. junija (Panška reka) oz. od 18. aprila do 18. julija (Menina planina), smo iz rastočih dreves odvzemali mikro izvrtke tkiv lesa, kambija in skorje. Z rotacijskim mikrotomom smo pripravili histološke rezine, jih obarvali z barviloma safranin in astra modro ter analizirali s svetlobnim mikroskopom in sistemom za analizo slike. Na mikroskopskih preparatih tkiv smo prešteli število slojev celic v kambijevi coni in merili sirino tekoče ksilemske branike. Dormantna kambijeva cona dreves z obeh rastišč je bila široka od 4 do 5 slojev celic. Pri prvih testnih drevesih s Panške reke so se celične delitve v kambijevi coni začele med 14. in 24. aprilom, na Menini planini pa med 9. in 16. majem. Na obeh rastiščih smo že teden dni po aktivaciji zasledili prve celice v fazi postkambijske rasti, sinteza sekundarne celične stene pa se je pričela teden do 2 kasneje. Prva dokončno diferencirana vlakna s popolnoma izoblikovano celično steno smo na obeh rastiščih zasledili 5-6 tednov po aktivaciji kambijeve cone. Kambijeva cona je bila na rastišču Panška reka najširsa (10 - 14 slojev celic) med 2. in 16. junijem, na Menini planini pa se število celic po aktivaciji ni bistveno spreminjalo in je variiralo med 8 in 10 celicami v radialnem nizu. Do konca eksperimenta nismo opazili novo nastalih celic kasnega lesa. Ugotovili smo, da so tako med drevesi in med rastiščema obstajale razlike v dinamiki nastajanja lesa in v širini ksilemske branike.

Secondary title: Beginning of cambial activity and early wood formation in beech (Fagus sylvatica L.) from two forest sites in growing season 2008
Secondary abstract: The beginning of seasonal activity in the cambial zone of beech (Fagus sylvatica L.) was investigated on 2 forest sites: Panška reka (400 m a.s.l.) and Menina planina (1,200 m a.s.l.). The beginning of cell differentiations and early wood formation in xylem tree-ring 2008 were observed. From each forest site, micro samples of wood, cambial zone and bark were removed from 6 living trees at weekly intervals, from March 17 to June 23 (Panška reka), and from April 18 to July 18 (Menina planina). Histological sections were prepared on a rotary microtome. They were stained with safranine and astra blue and analysed with a light microscope and with an image analysis system. On microscopic slides of tissues the cells in cambial zone were counted, and the widths of current xylem increment measured. The dormant cambial zone contained 4-5 layers of cells. In Panška reka the first divisions in the cambial zone began from April 14-24, on Menina planina from May 9-16. On both sites the first cells in post cambial growth were observed 1 week after activation of cambial zone, while secondary wall formation began 2 weeks later. The first completely differentiated fibres with completely formed cell walls were observed on both plots 5-6 weeks after activation of cambial zone. The widest cambial zone (10-14 cell layers) on Panška reka was observed from June 2-16. On Menina planina the cambial zone was constant after activation; it was from 8-10 cell layers wide, measured in radial direction. At the end of experiment no cells of late wood were tracked. Differences in wood formation dynamics and width of xylem tree-rings were found among trees and forest sites.
