Abstract

Glive predstavljajo dragocen vir prehranskih dopolnil, aminokislin, beljakovin, vitaminov, mineralov, ogljikovih hidratov in maščob. Proizvajajo številne bioaktivne spojine, ki jih lahko uporabljamo v industriji, biotehnologiji, farmaciji in medicini. Terapevtski gobi Ganoderma lucidum in Pleurotus ostreatus se že vrsto let uporabljata v zdravilne namene. Cilj magistrske naloge je bil optimizirati pogoje rasti micelarnih struktur (MS), kot so pH, čas gojenja in vrsta hranilnega tekočega medija, saj ti pogoji bistveno vplivajo na rast micelija in izoblikovanje MS. Najvišjo prirast MS G. lucidum smo dosegli v hranilnem mediju z dodatkom sladnega ekstrakta, medtem ko za P. ostreatus v mediju z dodatkom glukoze. Ugotovili smo, da je bila pri obeh glivah zagotovljena najboljša rast pri pH 8,5. Nadalje smo z optimiranjem časa ugotovili, da sta za nastanek MS G. lucidum potrebna 2 tedna, medtem ko za P. ostreatus 3 tedni. MS predstavljajo naravne, trajnostne in biorazgradljive materiale, ki se lahko uporabljajo tudi za terapevtske namene, kot je izdelava obližev za celjenje ran, saj se v njihovo strukturo učinkovito ujamejo različne učinkovine s protimikrobnimi lastnostmi. Proučevali smo morfologijo, kemijsko sestavo in hidrodinamične lastnosti pridobljenih MS. MS smo tudi funkcionalizirali z ultrazvočnim ekstraktom iz olupkov manga oz. antibiotikom ciprofloksacinom kot referenčno učinkovino. Primerjali smo njihove karakteristike z referenčnimi strukturami (MS brez funkcionalizacije z učinkovinami) ter določili učinkovitost ujetja, in vitro sproščanje ter protimikrobne lastnosti funkcionaliziranih MS. Rezultati kažejo na učinkovito ujetje učinkovin v MS obeh terapevtskih gob in postopno sproščanje učinkovin iz MS. Prav tako so MS z ekstraktom oz. antibiotikom uspešno zavirale rast Gram-negativnih (Escherichia coli, Pseudomonas aeruginosa) in Gram-pozitivnih bakterij (Bacillus cereus, Staphylococcus aureus).

Secondary title: Antimicrobial activity of functionalized micellar structures for therapeutic purposes
Secondary abstract: Mushrooms are a valuable source of nutritional supplements, amino acids, proteins, vitamins, minerals, carbohydrates and fats. They produce numerous bioactive compounds that can be used in industry, biotechnology, pharmacy and medicine. The medicinal mushrooms Ganoderma lucidum and Pleurotus ostreatus have been used for medicinal purposes for many years. The aim of this master thesis was to optimize the growth conditions of micellar structures (MS), such as pH, cultivation time and type of liquid culture medium, as these conditions significantly influence mycelial growth and MS formation. The highest MS yield of G. lucidum was obtained in a culture medium enriched with malt extract, while for P. ostreatus it was in a medium enriched with glucose. We found that the best growth for both fungi was ensured at a pH of 8.5. By optimizing the cultivation time, we also found that 2 weeks were required for the formation of MS in G. lucidum and 3 weeks in P. ostreatus. MS represent natural, sustainable and biodegradable materials that can also be used for therapeutic purposes, such as the production of wound healing patches, as various active ingredients with antimicrobial properties can be effectively incorporated into their structure. We investigated the morphology, chemical composition and hydrodynamic properties of the obtained MS. We also functionalized MS with an ultrasound extract of mango peel or the antibiotic ciprofloxacin as a reference drug. We compared their properties with reference structures (MS without drug functionalization) and determined the efficiency of incorporation, in vitro release and antimicrobial properties of the functionalized MS. The results indicate an efficient incorporation of active compounds into the MS of the two therapeutic fungi and a gradual release of the active compounds from the MS. In addition, the MS with the extract or the antibiotic successfully inhibited the growth of Gram-negative (Escherichia coli, Pseudomonas aeruginosa) and Gram-positive bacteria (Bacillus cereus, Staphylococcus aureus).
