Abstract

Diplomsko delo prikazuje razvoja nove stacionarne faze za izolacijo šestih elagitaninov (ET), galne (GK) in elagične kisline (EK) iz vodnega izvlečka kostanjevega lesa, z uporabo treh različnih polimerov, v vlogi stacionarne faze. Polimere, poli (stiren-ko-divinilbenzen) (PS/DVB), poliakrilat (PA) ter poli (vinilpirolidon-ko-divinilbenzen) (PVP/DVB) smo polimerizirali s suspenzijsko polimerizacijo, z uporabo iniciatorja α,α'-azobisisobutironitrila (AIBN) in stabilizatorjema polivinilpirolidonom (PVP) ter polivinil alkoholom (PVA). S temi polimeri smo napolnili v kromatografsko kolono za hitro kromatografijo ter preverili njihovo učinkovitost za ločevanja ET z uporabo preparativnega HPLC. Uspešnost ločbe smo preverili z analiznim HPLC. Pri tem smo spreminjanjali deleža acetonitrila (ACN) v mobilni fazi, dokler nismo dobili uspešne izolacije ET, GK in EK. Pogosto se je zgodilo, da so se vse komponente kostanjevega izvlečka izločile iz kolone v mrtvem času, ne glede na vrsto uporabljene stacionarne faze ali gradienta. Za takšno stacionarno fazo smo ocenili, da ni ustrezna za ločevanje danih spojin. Pri ločbi se je najbolj izkazala stacionarna faza PVP/DVB. To stacioanrno fazo smo nato uporabili za razvoj metode, s katero smo poskušali dobiti čim boljšo ločbo posameznih elagitaninov. Poleg tega smo opazovali tudi, kako na uspešnost ločbe vpliva povprečna velikost delcev stacionarne faze.

Secondary title: Development of a stationary phase for the separation of natural compounds
Secondary abstract: This thesis presents a study on the development of a new stationary phase for the isolation of six ellagitannins (ET), gallic acid (GK) and ellagic acid (EC) from an aqueous extract of chestnut wood, using three different polymers as stationary phases. The polymers, poly(styrene-co-divinylbenzene) (PS/DVB), polyacrylate (PA) and poly(vinylpyrrolidone-co-divinylbenzene) (PVP/DVB), were polymerised by suspension polymerisation using the initiator α, α'-azobisisobutyronitrile (AIBN) and the stabilisers poly(vinylpyrrolidone) (PVP) and poly(vinyl alcohol) (PVA). These polymers were loaded into a flash chromatography column, and their efficiency for ET separation was checked using preparative HPLC. The separation performance was verified by analytical HPLC. The proportions of acetonitrile (ACN) in the mobile phase were varied until we obtained a successful isolation of ET, GK and EK. It was often the case that all the components of the chestnut extract were eluted from the column in dead volume, regardless of the type of stationary phase used. Such a stationary phase was considered unsuitable for the separation of the compounds in question. The PVP/DVB stationary phase proved to be the most suitable for the separation. This stationary phase was then used to develop a method to obtain the best possible separation of the individual ellagitannins. In addition, we also observed how the average particle size of the stationary phase affects the success of the separation.
