Abstract

Stabilnost encimov je ključnega pomena pri njihovih različnih aplikacijah. Kot biokatalizatorje jih uporabljamo na različnih področjih kot so predelava hrana, okoljevarstvo in drugih industrijskih procesih. Encim β-laktamazo proizvajajo določene bakterije za razgradnjo β-laktamskega obroča v antibiotikih. Encim β-laktamazo lahko uporabljamo za razgradnjo antibiotikov in kot biomarker ugotavljanja ostankov antibiotikov v procesu čiščenja odpadnih voda. V diplomskem delu smo proučili vpliv magnetnega polja na specifično aktivnost in stabilnost encima β-laktamaze v praškasti obliki in v vodni raztopini, in sicer smo encim izpostavili konstantnemu in alternirajočemu (sinusnemu) magnetnemu polju ter spreminjali čas izpostavitve. Tako smo proučevali stabilnost β-laktamaze v prahu in v vodni raztopini pri danih pogojih. Po izpostavitvi vzorcev magnetnemu polju smo z encimskim testom določili preostalo aktivnost encima in koncentracijo proteinov ter tako določili vpliv magnetnega polja na stabilnost β-laktamaze.

Secondary abstract: The stability of enzymes is crucial in various applications. As biocatalysts, they are used in analytical chemistry, food processing, environmental protection and industrial processes. The enzyme β-lactamase is produced by bacteria to break down the β-lactam ring in antibiotics, allowing them to survive. The enzyme can be used to detect and remove the remains of antibiotics in wastewater treatment processes. In this thesis, the effect of magnetic field on the specific activity and stability of the β-lactamase enzyme was studied by exposing the enzyme to constant and alternating (sinusoidal) magnetic fields with varying time of exposure. The stability of β-lactamase was studied under the given conditions. After exposure of the samples to the magnetic field, the differences in specific activity and protein concentration were monitored using the enzyme activity assay and Bradford method for determination of total protein content.
