Secondary title: Speciation within the species complex of the Grayling (Hipparchia semele) on the Balkan Peninsula
Secondary abstract: In this thesis we wanted to determine whether the butterfly species complex within the genus Hipparchia differ on the basis of genetic markers and the shape of their genital apparatus. By using the phylogenetic analysis, we checked the genetic relatedness between individuals based on mitochondrial COI gene and the nuclear gene GADPH. Species of the genus Hipparchia are morphologically very similar they differ only in the anatomy of the male genitalia. We cut the last part of the abdomen of the sampled males and prepared permanent preparations according to the protocol. Under the microscope, we measured the length of selected genital parts and analysed them. Mitochondrial COI gene was suitable for separating H. senthes from other species, but not of the H. semele from H. volgensis. The nuclear gene GAPDH is unsuitable for identification the studied species. Male genitalia analysis proved to be appropriate for determining the butterflies of the genus Hipparchia. Genitalia of the H. semele and H. volgensis are generally similar they differ only in the length and width of the valve. H. senthes has significantly smaller genitalia compared to the other two studied species. It is probable that incompatibility based on the size of genitalia is a major factor in the development of mechanisms of reproductive isolation among species in the H. semele complex.
