Abstract

V diplomskem seminarju obravnavamo pojav aspirinsko inducirane astme s pomočjo fizikalno-matematičnega modela presnove arahidonske kisline. Že obstoječi model nadgradimo in na podlagi razlik v ekspresiji encimov prostaglandin E sintaze (PGES) in levkotrien C4 sintaze (LTC4S) identificiramo aspirinsko intolerantne in tolerantne astmatike ter neastmatike. Modelni rezultati napovejo največje tveganje bronhokonstrikcije po zaužitju NSAR pri populaciji s povišano ekspresijo encima LTC4S in znižano ekspresijo PGES. Modelne napovedi primerjamo z izmerjenimi razmerji koncentracij med posameznimi eikozanoidi in ugotavljamo zelo dobro ujemanje modela z meritvami iz dveh različnih virov.

Secondary abstract: In the graduate seminar we discuss the occurrence of aspirin-induced asthma using a mathematical model of arachidonic acid metabolism. We upgrade the existing model and based on the differences in the expressions of the enzymes prostaglandin E synthase (PGES) and leukotriene C4 synthase (LTC4S) we identify aspirin intolerant and tolerant asthmatics as well as nonasthmatics. The model predicts the highest risk of bronchoconstriction in the population in which the expression of enzyme LTC4S is increased and expression of PGES is decreased. The model results are in good agreement with the measured concentration ratios between particular eicosanoids, obtained from two different experimental studies.
