Abstract
Diplomsko delo opisuje sintezo encimskih skupkov iz encima celulaza ali na kratko CLEAs ter vpliv superkritičnega ogljikovega dioksida (SC CO2) na aktivnost teh skupkov. Priprava zamreženih encimskih skupkov poteka po določenem postopku, katerega ključna dela sta obarjanje in zamreženje. Torej obarjanje topnega encima z ustreznim organskim topilom in nadaljnjo zamreženje tako oborjenega encima s pomočjo mrežnega povezovalca. V našem primeru smo kot mrežni povezovalec uporabili glutaraldehid in aceton kot obarjalni reagent. Zamrežene encimske skupke (CLEAs) smo izpostavili pri temperaturah 35 °C, 40 °C, 50 °C in 60 °C. Istočasno smo jih izpostavili v SC CO2 v visokotlačnem šaržnem reaktorju, in sicer pri temperaturah 35 °C, 40 °C, 50 °C in 60 °C in tlaku 100 bar ter pri temperaturah 35 °C in 40 °C ter tlaku 200 bar. Aktivnost CLEAs je s podaljšanjem inkubacijskega časa upadala.

This undergraduate thesis examines the synthesis of cross-linked enzyme aggregates or CLEAs from cellulase and the impact of supercritical carbon dioxide (SC CO2) on the activity of these aggregates. Cross-linked enzyme aggregates are produced using a special procedure composed of two key stages: precipitation and cross-linking. The soluble enzyme is precipitated with a suitable organic solvent and the enzyme precipitated in this manner is subsequently cross-linked using a cross-linking agent. In this study, acetone was used as the precipitant and glutaraldehyde as the cross-linker. CLEAs were exposed to atmospheric pressure at various temperatures (35 °C, 40 °C, 50 °C, and 60 °C). They were also exposed to SC CO2 in a high-pressure batch reactor at 35 °C, 40 °C, 50 °C, and 60 °C and 100-bar pressure, and at 35°C and 40 °C and 200-bar pressure. Exposing the CLEAs to various temperatures and pressures for various periods of time revealed that the activity of aggregates decreases with time. |
