Secondary abstract: | The aim of this graduate thesis was to study the influence of reaction parameters (temperature, time and concentration) on the hydrolytic ability of subcritical water on the basic example of diglicoside quercetin – rutin. Highest yield of quercetin was based on experimental design and response surface methodology optimal reaction parameters at wich the highest yield of quercetin is obtained were determined. All experiments were performed in a high-temperature high-pressure autoclave using an inert (nitrogen) atmosphere at condition, which were predefined using Design-Expert 7.0 software. Yield of quercetin and other products of hydrothermal degradation in samples after treatment with subcritical water were determined using high-performance liquid chromatography. Obtained results were optimized using response surface methodology and the optimal parameters, which contributed to the highest yield of quercetin were determined. |
