Abstract
Prevladujoča skupina gliv v solinah Jadranskega morja so črne kvasovke. Ena izmed njih je Phaeotheca triangularis, ki smo jo v tem diplomskem delu obravnavali. P. triangularis je halofilna kvasovka, ki v solinah najbolje uspeva pri slanosti od 22 do 28 %. Celice P. triangularis smo mehansko razbijali s homogenizatorjem od 10 do 90 min pri atm. tlaku in temperaturi 35 °C. Za primerjavo smo celično suspenzijo P. triangularis inkubirali pri atm. tlaku in temperaturi 35 °C. S homogenizatorjem so se celične stene kvasovk razbile in njihova vsebina se je izlila v tekoč medij. Tako smo lahko preučevali vpliv homogeniziranja na izločanje encimov in proteinov iz celične suspenzije P. triangularis v odvisnosti od časa izpostavitve. Določevali smo preostalo aktivnost encimov celulaze, β-glukozidaze, α-amilaze in proteaze tako, da smo na UV-Vis spektrofotometru merili absorbance supernatantov pri različnih valovnih dolžinah. Zanimal nas je vpliv mehanskih sil na preživelost celic P. triangularis v celični suspenziji. Ugotovili smo, da je preživelost celic P. triangularis s podaljševanjem časa homogenizacije upadala, medtem ko je koncentracija proteinov v celični suspenziji P. triangularis naraščala.

The dominant group od fungi in salterns of the Adriatic Sea are black yeasts. One of them is Phaeotheca triangularis, which is discussed in this diploma work. P. triangularis is halophilic yeast that groves in the salterns best in salinity from 22 to 28%. The cells of P. triangularis were mechanically disrupted by using the homogeniser from 10 to 90 min at atmospheric pressure and temperature 35 °C. For comparison, a cell suspension of P. triangularis was incubated at atmospheric pressure and temperature 35 °C. The yeasts cell walls were disrupted and their content was spilled into the liquid medium. The impact of homogenization on secretion of enzymes and proteins from cell suspension P. triangularis was studied. The residual activity of the enzymes cellulase, β-glucosidase, α-amylase and protease was studied by measuring absorbance of the supernatants at different wavelengths on a UV-Vis spectrophotometer. The influence of mechanical forces on the survival of cells of P. triangularis in the cell suspension was a subject of interest. The survival of cells of P. triangularis was decreasing with the time of homogenization, while the concentration of the proteins in the cell suspension of P. triangularis was increasing. |
