Abstract

Namen diplomskega dela je bila optimizacija analizne metode po ekstrakciji, s katero smo določili vsebnost kloridnih, nitratnih in oksalatnih anionov v ekstraktu tara (Colocasia esculenta (L.) Schott)) s pomočjo ionske kromatografije (IC). Ekstrakcijo smo optimirali glede: načina izvedbe, temperature in učinkovitosti Tween-reagenta. Za realni vzorec, ekstrakt tara, smo študirali sestavo mobilne faze in sicer tako, da smo dobili zadovoljivo ločljivost pikov, ki jih želimo kvantitativno ovrednotiti. Analizno metodo smo tudi delno validirali, saj smo določili: linearnost, mejo zaznavnosti, mejo določljivosti, natančnost, pravilnost, vpliv matrice in časovno stabilnost standardov. V realnih vzorcih tara, smo določili vsebnost kloridnih, nitratnih in oksalatnih ionov v koncentracijskem območju od 197 mg/kg do 2 930 mg/kg SS, od 11 mg/kg do 10 868 mg/kg SS in od 761 mg/kg do 2 680 mg/kg SS.

Secondary abstract: The purpose of the diploma thesis was the optimisation of analytical method, for the determination of chloride, nitrate and oxalate anions content in taro (Colocasia esculenta (L.) Schott) extracts usng ion chromatography (IC). The extraction was optimised with regard to manner of performance and extraction temperature. Teh efficiency of Tween 80-reagent for the extraction of nitrate ions was also studied. We optimised the composition of the mobile phase for a real extract of taro sample in order to obtain complete separation od chromatographic peaks for anions of interest. The analytical method was also partly validated. We determined: linearity, limit of detection and quantification, precision, trueness, matrix influence and temporal stability of standard solutions. The contents of chloride, nitrate and oxalate ions were in the ranges from 197 mg/kg to 2 930 mg/kg DM, 11 mg/kg to 10 868 mg/kg DM and 761 mg/kg to 2 680 mg/kg DM, respectively
