Abstract
Hitozan je zaradi svoje dokazane varne uporabe primeren tudi za razvoj novih protimikrobnih tekstilnih sistemov, kljub temu pa mu ni moč pripisati lastnosti antioksidativnosti, ki lahko pripomore k hitrejšemu celjenju ran ali razjed. Zaradi tega je smiselno v sistem s hitozanom vključiti katero izmed naravnih spojin z dokazanim antioksidativnim delovanjem. Tem kriterijem ustreza eugenol. Namen tega magistrskega dela je bil razviti formulacijo hitozana v kombinaciji z eugenolom v različnih strukturnih oblikah (raztopina, nanodelci, oborina) za funkcionalizacijo viskoze. Protimikrobna in antioksidativna formulacija bi naj služila kot funkcionalen premaz, prednostno na področju celjenja ran na koži in sluznici v ustni votlini. S stališča različnih strukturnih oblik, ki so ključnega pomena za dostopnost protimikrobnih aminskih skupin in za delež vezanega eugenola v sistem s hitozanom, smo predvideli, da bodo posamezne strukturne oblike različno vplivale na ciljani lastnosti protimikrobnost in antioksidatovnost. V prvi fazi je bila izvedena sinteza nanodelcev (hitozana in eugenola, kapsuliranega v hitozan), ki so bili okarakterizirani z vidika morfologije (SEM) ter velikosti in porazdelitve nanodelcev (DLS). Po opravljeni funkcionalizaciji viskoze s posameznimi strukturnimi oblikami obdelovalne kopeli je bila uporabljena FT-IR spektroskopija za proučitev tipičnih funkcionalnih skupin, potenciometrična titracija, podprta z metodo Acid Orange VII, za proučitev deleža aminskih skupin, ter vrednotenje mikrobioloških lastnosti in antioksidativnega značaja funkcionalizirane viskoze. Desorpcijska kopel je bila ovrednotena na osnovi merjenja pH vodnega ekstrakta. Preostali delež vezanega hitozana po desorpciji je bil, enako kot v primeru funkcionalizacije, proučen z metodo Acid Orange VII. Ugotovljeno je, da različne strukturne oblike hitozana in hitozana v kombinaciji z eugenolom različno vplivajo na protimikrobnost in antioksidativnost funkcionalizirane viskoze. Protimikrobna in antioksidativna formulacija se je izkazala za primeren funkcionalen premaz za razvoj tekstilij za potencialno uporabo v medicini, prednostno na področju celjenja ran ter sluznic.

Chitosan is proven to be safe for use, and is therefore suitable for the development of new antimicrobial textile systems. However, antioxidant properties that can help e.g. in wounds or ulcers healing, cannot be attributed to chitosan. For this reason, it makes sense to include any of the natural compounds with proven antioxidant action in a system with chitosan. These criteria are fulfilled by eugenol. The purpose of this master's thesis was to develop an additive formulation of chitosan in combination with eugenol in different structural forms (solution, nanoparticles and precipitation) for viscose functionalization. Antimicrobial and antioxidant formulation should serve as a functional coating, preferably in the field of wound healing of the skin and healing of ulcers of the oral cavity. From the perspective of different structural forms that are crucial for the availability of antimicrobial amine groups and the proportion of eugenol, bound in the system with chitosan, it was assumed that individual structural forms would have a different impact on the targeted properties, i.e. antimicrobial and anti-oxidative ones. The first stage was synthesis of nanoparticles (chitosan, as well as eugenol encapsulated in chitosan), which were characterized from the point of view of morphology (SEM), and of the size together with the distribution of nanoparticles (DLS). After functionalization of viscose, FT-IR spectroscopy was used to examine typical functional groups, potentiometric titration to examine the proportion of amino groups, it was supported by the method of Acid Orange VII, and evaluation of microbiological properties together with anti-oxidative activity. Desorption bath was assessed by measuring the pH of aqueous extract. The remaining chitosan bound after desorption was examined by the method of Acid Orange VII, as in the case of functionalization. It was proven that various structural forms of chitosan and chitosan in combination with eugenol differently affect antioxidant and antimicrobial properties of functionalized viscose. Antimicrobial and antioxidant formulation has proven to be a suitable functional coating for the development of textiles for potential use in medicine, preferably in the field of wound and oral mucosa healing. |
