Abstract
V diplomskem delu sem predstavila pomen ustne higiene za nastajanje zobnega plaka in s tem povezano teorijo temeljnega kamna predvsem pri razvoju parodontoze. Poudarek naloge je na lastnostih bakterije Aggregatibacter actinomycetemcomitans in toksinu Cdt, ki ga omenjena bakterija sintetizira. Namen diplomske naloge je bil sprožiti sintezo različice rekombinantnega toksina CdtB in preveriti vpliv toksina na bakterijsko rast. Za cilj smo si zadali izvedbo testa za analizo vpliva različice toksina CdtB na preživelost producentske bakterije Escherichia coli. Uporabili smo metodo verižne reakcije s polimerazo in druge osnovne molekularno biološke tehnike. Dokazali smo, da–posamezna varianta toksina CdtB različno vpliva na rast producentskih bakterij in da različica in ne divji tip toksina Cdt prepreči rast bakterije Escherichia coli.

In my diploma I am presenting oral hygiene, dental plaque formation and in association with that, keystone pathogen hypothesis for periodontitis development. The characteristics of bacterium Aggregatibacter actinomycetemcomitans, which is strongly connected to periodontal disease and its Cdt toxin are described. The purpose of the diploma, was to determine the effect of the synthesis of recombinant Cdt toxinand its variant on the growth of the producing bacteria. The aim was to implement a test to analyze the effect of recombinant CdtB toxin variants on the survival of the producing Escherichia coli. To accomplish this, we applied basic molecular-biology techniques. We showed that we can discriminate between the CdtB toxin according to the count of the colony forming, and that the toxin variant, but not the wild type CdtB toxin prevent growth of the Escherichia coli. |
