Abstract

Ekstrakcija z organskim topilom je razširjena metoda za izolacijo zdravilne učinkovine iz fermentacijske brozge. Določitev optimalnih pogojev ekstrakcije je pomembna za optimizacijo izkoristka, ter izdelavo masne bilance, ki je osnova za projektiranje proizvodne linije. Cilj tega diplomskega dela je bil določiti optimalne pogoje enostopenjske ekstrakcije makrolida iz fermentacijske brozge s toluenom. Optimalni čas ekstrakcije je bil predhodno že raziskan, in sicer znaša 2 uri. V sklopu eksperimentalnega dela diplomske naloge smo določili vpliv temperature in pH na ekstrakcijo makrolid laktamov iz fermentacijske brozge s toluenom, pri čemer smo ugotovili, da so optimalni pogoji ekstrakcije pri temperaturi 45°C in pH 4,0. Ugotovili smo, da se izkoristek ekstrakcije poveča, pri čemer se zmožnost ločbe faz po obdelavi fermentacijske brozge s homogenizatorjem lahko zmanjša. Vzrok za slabšo ločbo faz so majhni delci, ki pri obdelavi s homogenizatorjem nastanejo iz delcev značilnih za bioprocese.

Secondary abstract: Extraction with an organic solvent is widely used method for isolating active substances from fermentation broth. Determination of optimal extraction conditions is important for optimizing efficiency and determining mass balance which is the basis for designing the production line. The aim of this study was to determine the optimal conditions for a single-stage extraction of macrolide from the fermentation broth with toluene. Optimal extraction time has previously been detrmined and is set to 2 hours. Aim of the experimental work was to determine the effect of temperature and pH on the process of extraction of macrolide-lactams from the fermentation broth with toluene. Optimal extraction conditions were set at a temperature of 45 ° C and pH 4.0. We have found that the yield of the extraction increases whereas the ability of phase separation after treating fermentation broth with homogenizer can decrease. The reason for the poor separation of phases are small particles, arising from bio processes particles, when treated whit homogenizer.
