Abstract

Namen diplomskega dela je razvoj in validacija metode B po standardu SIST EN ISO 7887:2012 Kakovost vode – Pregled in določevanje vode. V laboratoriju NLZOH Novo mesto smo se glede na spremembe zahtev zakonodaje, naročnikov in akreditacijske presoje odločili izvesti ponovno validacijo, saj se z leti parametri spreminjajo. Dodali so standard za preverjanje točnosti valovnih dolžin, primerjali so pH in barvo in pregledali vpliv interferenc. Določili smo LOD in LOQ, delovno območje, povprečje, standardno deviacijo in RSD. Preverili smo pravilnost in natančnost metode ter določili merilno negotovost. Metoda se uporablja za vse tipe vod naravne in odpadne vode. Pri realnih vzorcih različnih tipov vod smo s spektrofotometrom Agilent 8453 UV/VIS izmerili absorpcijo pri treh različnih valovnih dolžinah 436 nm, 525 nm in 620 nm, pri katerih smo izračunali realno barvo vode. Glede na rezultate validacije smo ocenili, da je metoda primerna za rutinsko določanje barve vode.

Secondary abstract: The purpose of this diploma thesis is to develop and validation the B method according to the standard SIST EN ISO 7887:2012 for determining water quality. In the NLZOH laboratory Novo mesto, we have decided to carry out the re-validation as the parameters change over the years. They added the standard for checking the accuracy of wavelengths, they are comparing pH and color and examined the influence of interference. We determined LOD and LOQ values and the operating limits. We checked accuracy and precision of the method and determined the uncertainty of measurement. The method is used for all types of water: natural waters and wastewaters. For real samples of differents types of waters, we used the spectrophotometer Agilent 8453 UV/VIS to measure absorbance at three different wavelengths: 436 nm, 525 nm and 620 nm in which we calculated the real color of the water. In the light of the results of validation we have assessed that the method is suitable for routine determination of water quality.
