Abstract

Rastlinski materiali vsebujejo veliko zdravilnih učinkovin in med njimi so tudi tiste, ki jih najdemo v eteričnih oljih. Najpomembnejše komponente v eteričnih oljih, ki jim dajejo značilen vonj so enostavni terpeni – monoterpeni. Za nas pomembna monoterpena sta karvon in limonen, ki se nahajata v semenih kopra in kumine ter listih zelene mete. Namen diplomske naloge je bil določiti optimalne ekstrakcijske parametre za izolacijo karvona in limonena iz semen kumine, kopra in listov klasaste zelene mete z različnimi ekstrakcijskimi postopki. Eterična olja smo izolirali s konvencionalno metodo destilacije z vodno paro po Clevenger-ju, z visokotlačnimi ekstrakcijami s superkritičnimi fluidi (CO2) v laboratorijskem merilu, z ekstrakcijo po Soxhletu in z ultrazvočno ekstrakcijo. Kvalitativno in kvantitativno sestavo olj smo določili s pomočjo plinske kromatografije z masno spektrometrijo. Komponenti, limonen in karvon smo identificirali s pomočjo ustreznih standardnih substanc in jih kvantitativno ovrednotili z metodo eksternega standarda s plinsko kromatografijo sklopljeno z masno spektrometrijo.

Secondary abstract: Plants are considered as an important source of active ingredients, used also for medicinal products. Many of them are found in essential oils. One of the main components of essential oils are simple terpenes, known as monoterpenes. Carvone and limonene are both monoterpenes, found in the dill seeds, caraway and in spearmint leaves. The aim of this thesis was to determine the optimal extraction parameters for isolation of carvone and limonene from dill seeds, caraway and leaves of spearmint using various extraction processes. Essential oils were first isolated by conventional method by steam distillation using Clevenger apparatus. The second approach of isolation was supercritical extraction, using supercritical carbon dioxide. Finally, Soxhlet extraction and ultrasonic extraction were also performed for comparison. Gas chromatography – mass spectroscopy was used in order to determine qualitative and quantitative composition of essential oils. External standard method was used for identification and quantification of limonene and carvone by gas chromatography – mass spectroscopy.
