Abstract
V diplomskem delu preučujemo delovanje modela sklopljenih celic beta. Te endokrine celice se nahajajo v Langerhansovih otočkih in izločajo inzulin ter s tem uravnavajo raven glukoze v krvi. Samo delovanje teh celic je zelo kompleksno in sestoji iz številnih med seboj povezanih dinamičnih procesov. Zavoljo boljšega razumevanja se pri preučevanju pogosto poslužuje modeliranja in preučevanja modelov. V nalogi je podrobneje opisana fiziologija Langerhansovih otočkov in pregled razvoja modelov celic beta. V nadaljevanju predstavimo eksperimentalne tehnike, ki se uporabljajo za snemanje aktivnosti celic beta v akutnih rezinah trebušne slinavke miši, in večcelični model, s katerim smo izvajali simulacije. Rezultati pokažejo, da napoved modela dobro sovpada z eksperimentalnimi opažanji, v kolikor se upošteva heterogenost celic beta in časovni zamik, ki je posledica metabolnih procesov v celicah.

In the thesis we study the functioning of a computational model of interconnected beta cells. These endocrine cells are located in islets of Langerhans and secrete insulin and thereby regulate the blood glucose level. The sole function of these cells is very complex and consists of many interconnected dynamic processes. Modeling and studying the models are often utilized for reasons of a better understanding on examining. In the thesis we describe the physiology of islet and provide an overview on the development and examination of beta cell models. Further we present experimental techniques used to record activity of beta cells in acute slices of the mouse pancreas, and a multicellular model on the basis of which we carried out simulations. The results show that the predictions of the model fit well with the experimental observations, if the heterogeneity of the beta cells and the time delay caused by the metabolic processes in cells are taken into the account. |
