Abstract

Namen naše raziskave je bila primerjava ultrastrukturnih značilnosti celic beta Langerhansovega otočka v odvisnosti od trajanja izpostavitve glukozi in različnih koncentracij glukoze. Želeli smo raziskati delovanje celic beta in dogajanja v zvezi z granulami inzulina v veziklih v opisanih razmerah. Pripravili smo tkivne vzorce in jih analizirali s pomočjo svetlobne mikroskopije in transmisijske elektronske mikroskopije (TEM). Dobljene podatke smo statistično ovrednotili. V raziskavi smo potrdili, da so za proučevanje vpliva koncentracije glukoze na celice beta vibratomske rezine, vključene v epoksidno smolo, učinkovitejše od raziskav na koščkih tkiva, vključenih v epoksidno smolo, ker se endokrino tkivo v raztopinah do 12 mM ohranja nepoškodovano vsaj eno uro. Tudi v tkivnih rezinah je eksokrini del po daljšem času poškodovan, tako da bi za raziskave na eksokrinem tkivu opisano metodo bilo potrebno optimizirati z namenom boljšega preživetja celic. V celicah beta nismo opazili jasnih in konsistentnih morfoloških sprememb, razen pri koncentraciji glukoze 12 mM, kjer je bila velikost veziklov in granul po 15 minutah manjša kot po 30 in 60 minutah. V prihodnjih raziskavah priporočamo uporabo večjega števila živali, otočkov in celic, optimizacijo metode za uporabo na acinarnem tkivu in uporabo na zdravem in bolnem tkivu živalskega in človeškega porekla.

Secondary abstract: The aim of the present study was comparison of ultrastructural features of insulin secreting beta cells exposed to different concentrations of glucose and different duration of exposure to it. We wanted to explore functioning of the beta cells in described conditions. After the preparation of tissue slices, we analyzed them with light microscopy and transmission electron microscopy (TEM). The obtained data were statistically evaluated. We confirmed that it is better using tissue slices when studying the impact of the concentration of glucose in the beta cells than tissue cubes. It is more effective because the endocrine tissue in glucose solution of 12 mM keeps intact for at least one hour. Even in tissue slices, after longer periods of time the exocrine tissue becomes too damaged to be studied and enables studies of acinar cells. The method should be optimized to ensure better viability. In beta cells, no clear and consistent changes were observed with respect to time of exposure and glucose concentration employed. Only with 12 mM glucose, all parameters of vesicle and granule size suggested a consistent increase in size from 15 minutes after exposure to 30 and 60 minutes after exposure. In future studies, we recommend the use of greater number of animals, islets, and cells. Moreover, the method should be optimized as to enable studying the exocrine part of the pancreas, and as well as normal also diseased tissue of animal and human origin could be used.
