Abstract

Namen diplomskega dela je bil validirati analizno metodo za kvalitativno in kvantitativno določanje antocianinov v rdečem sadju in sadnih sokovih iz rdečega sadja. S tem smo želeli izboljšati metodologijo kontrole kvalitete naravnih produktov, ki vsebujejo naravna barvila (EEC E163). Z visokotlačno tekočinsko kromatografijo (HPLC) smo analizirali metanolne ekstrakte rdečega sadja v 6 vzorcih slovenskega porekla in sadnih sokov iz rdečega sadja, v 16 vzorcih slovenskega porekla in za primerjavo še 3 vzorce iz tujine (Avstrija in Italija). Za potrebe razvrščanja je bila prvotno potrebna predhodna identifikacija značilnih antocianinov za posamezno vrsto rdečega sadja s pomočjo standardnih raztopin dostopnih antocianinov. Opravili smo validacijo analizne metode, določili linearnost, natančnost, točnost, mejo določanja in časovno stabilnost metanolnega ekstrakta. Na osnovi vseh rezultatov smo ugotovili, da gre v Sloveniji za kvalitetne izdelke rdečega sadja in sadnih sokov iz rdečega sadja, saj se nikjer med analiziranimi vzorci ne pojavljajo signifikantne razlike.

Secondary title: Characterization of anthocyanins profiles in red fruits and red fruits juices
Secondary abstract: The purpose of this thesis was to validate the analytical method for qualitative and quantitative determination of anthocyanins in red fruits and red fruits juices in order to improve the quality control methodology of natural products containing natural colourants (EEC E163). High performance liquid chromatography was used for determination of anthocyanins in methanol extracts of red fruits in 6 samples of Slovenian origin and red fruits juices in 16 samples of Slovenian origin. For comparison, we have determined the anthocyanins content in 3 other samples originating from Austria and Italy. The approach was based on previous identification of typical anthocyanins profiles of each - red fruits using standard solution of available anthocyanins. The analytical method has been validated and the linearity, precision, accuracy, limit of quantification and stability of methanol extracts were determined. Based on all final results we have concluded that the quality of Slovenian products of red fruits is satisfactory, because there were no significant differences between the analysed samples.
