Povzetek

Razvili smo elektrokemijsko celico, ki služi kot modelni biosenzor, ter kot aktivno komponento vsebuje kvasovke vrste Saccharomyces cerevisiae. Preizkušali smo različne postopke sestave elektrokemijskih celic. Pri postopku sestave elektrokemijske celice smo uporabili nerjavno jeklo, površina kovinske elektrode, pa je bila po potrebi modificirana s tanko plastjo volframovega trioksida. Na delovno elektrodo smo kot biološko komponento nanašali kvasovke Saccharomyces cerevisiae. Z modelnim biosenzorjem smo nato spremljali, kako se ob spreminjanju različnih parametrov, le-ta odziva na 10 % raztopino metanola. Za detekcijo smo uporabili metodo elektrokemijske impedančne spektroskopije (EIS), s katero smo merili frekvenčno odzivnost električnega toka in s tem pridobili podatke o plasti kvasovk na površinah elektrod, kot rezultat pa smo interpretirali Bodejeve in Nyquistove diagrame. Stabilnost naprave in pripravljenih plasti smo predhodno določevali z merjenjem potenciala odprtega tokokroga (OCP). Topografijo in sestavo elektrod smo dodatno preverjali s tehniko vrstične elektronske mikroskopije (SEM) in optično mikroskopijo. Ugotovili smo, da je pri izdelavi biosenzorja izrednega pomena debelina nanosa biološke komponente, postavitev in velikost elektrod ter izbira materiala. Dokazali smo, da so preiskani elektrokemijski biosenzorji učinkoviti pri detekciji metanola v vodnih raztopinah. Razviti modelni biosenzorji so cenovno ugodni in nam služijo kot študijski primer za razvoj kompleksnejših biosenzorjev, ki kot aktivno plast uporabljajo žive celice.

Sekundarni povzetek: An electrochemical cell that serves as a model biosensor and contains yeast Saccharomyces cerevisiae as the active component was developed. Different configurations of electrochemical cells were assembled and tested. Stainless steel was used in the electrochemical cell composition process and the surface of the metal electrode was modified with a thin layer of tungsten trioxide if necessary. The yeast Saccharomyces cerevisiae was applied to the working electrode as a biological component. The model biosensor was then used to monitor its respond to a 10 % methanol solution when changing various parameters. For detection of the frequency response of the electric current method of the electrochemical impedance spectroscopy (EIS) was applied. The data on the yeast layer of the electrode surfaces was obtained with portable potentiostat/galvanostat using EIS method, and as a result the Bode and Nyquist plots were interpreted. The stability of the device and the prepared layers were previously determined by measuring the potential of the open circuit (OCP). The topography and composition of the electrodes were additionally verified by the technique of scanning electron microscopy (SEM) and optical microscopy. It was observed that thickness of the biological component layer, the layout of the electrodes, the size of the electrodes and the choice of the material, are of paramount importance for the biosensor assembly. It was demonstrated that the electrochemical biosensor is effective in the detection of methanol in aqueous solutions. The model biosensor serves as a case study for the development of more complex biosensors that utilize living cells as their active layer.
