Povzetek
Magistrsko delo prikazuje izvedbo delne validacije metode za določanje As, Ba, Cd, Co, Cr, Cu, Mn, Mo, Ni, Pb, Se, Sr in Zn v vzorcih mesa, mesnih izdelkov in morske hrane z uporabo induktivno sklopljene plazme z optično emisijsko spektroskopijo, pripravo in mikrovalovni kislinski razklop vzorcev ter določanje izbranih kovin v njih. Za vsako testirano kovino smo določili meji zaznavnosti in določljivosti. Z izračunom kvadratov korelacijskih koeficientov umeritvenih premic, večjih od 0,990, smo potrdili linearnost v izbranem koncentracijskem območju. Z izračuni izkoristkov meritev raztopin standarda znane koncentracije, ki so bili znotraj kriterija med 80 % in 120 %, smo potrdili točnost. Točnost smo nato preverili še s testiranjem izkoristkov realnega vzorca, ki smo mu dodali znano količino raztopine standarda, in jo potrdili za večino kovin, razen za Sr. Z izračuni relativnih standardnih odmikov meritev raztopine standarda, manjših od 20 %, smo potrdili natančnost (ponovljivost). Opazovali smo, kako se signal ICP-OES instrumenta spreminja s številom injiciranj ter določili, po koliko injiciranjih je potrebno ponovno umerjanje instrumenta. V vseh vzorcih smo določili kakšno izmed testiranih kovin, razen Pb in Se. Izračunali smo, koliko g/dan vzorcev, v katerih smo določili prisotnost kovin, bi povprečna odrasla oseba lahko pojedla, da še ne bi presegla postavljenih mej za vnos kovin v telo. Te količine smo primerjali s porcijami podobnih izdelkov iz raziskav. Povprečna odrasla oseba bi z uživanjem škampov, školjk dagenj in govejih jeter v poročanih količinah presegla nekatere meje za vnos As in/ali Cd.

Determination of heavy metals in meat, meat products and seafood by ICP-OES |

This master’s thesis presents partial validation of the method for the determination of As, Ba, Cd, Co, Cr, Cu, Mn, Mo, Ni, Pb, Se, Sr, Zn in meat, meat products and seafood, using inductively coupled plasma with optical emission spectroscopy, preparation and acid-assisted microwave digestion of samples and determination of selected metals in these samples. For each of the selected metals, we determined limits of detection and quantification. By calculating the squares of the correlation coefficients of the calibration lines, greater than 0,990, we confirmed the linearity within the selected concentration range. We confirmed the accuracy by calculating the recoveries of the measurements of the solution of standard with known concentration, which were within the criterion between 80 % and 120 %. We then checked the accuracy by testing a real sample spiked with a known amount of the solution of standard and we confirmed it for most metals, except for Sr. We confirmed the precision (as repeatability) by calculations of the relative standard deviations of the measurements of the solution of standard, smaller than 20 %. We were observing how the ICP-OES signal of the instrument changes with the number of injections, and we determined after how many injections the instrument needs to be recalibrated. We determined at least one of the metals in all samples, except for Pb and Se. For the samples in which the metals were determined, we calculated how many g/day of these products an average adult could eat to not exceed the set limits for the intake of metals into the body. We compared these amounts with portions of similar products from studies. By consuming samples of shrimp, mussel shells and beef liver in the indicated daily portions from research, the average adult would exceed some of the established limits for the permissible intake of As and/or Cd. |
