Povzetek

Scoliopteryx libatryx uvrščamo v družino Erebidae in naddružino Noctuoidea. Obstajata dve generaciji, druga pogosto prezimuje v diapavzi v vhodnih delih podzemeljskih jam. Zanimalo nas je, kako se spreminjajo masa, zaloge lipidov in glikogena ter vsebnost vode v osebkih med prezimovanjem ter ali obstajajo razlike med spoloma. Zanimalo nas je tudi, katere spremembe v ultrastrukturi se v tem obdobju naravnega stradanja dogajajo v epitelnih celicah Malpighijevih cevk (MC). Raziskave smo opravili z optičnim in elektronskim mikroskopom ter z ustreznimi biokemijskimi analizami.Ugotovili smo, da so pri S. libatryx glavne energijsko-založne snovi v času prezimovanja lipidi. MC so sestavljene iz enoslojnega epitela, bazalne lamine in posameznih mišičnih celic. Enoslojni epitel sestavljajo primarne epitelne celice, ki so na bazalnem delu s hemidezmosomi povezane z bazalno lamino. Med uvihki bazalne plazmaleme so mitohondriji. Jedra so pri večini celic v bazalnem delu. V jedrih je heterokromatin, okoli jedra pa obilen zrnati endoplazmatski retikulum. Perinuklearni del je izpolnjen s sferiti. Pod apikalno membrano je aktinski korteks s številnimi mitohondriji. Apikalna membrana je izdiferencirana v številne mikrovile, v katerih so pogosto mitohondriji. Mikrovili močno povečajo površino, skozi katero poteka izločanje snovi v lumen MC. Med prezimovanjem se vsebina sferitov postopoma izprazni, obenem pa se pojavljajo avtofagne structure, vključno z rezidualnimi telesi. Med spoloma nismo odkrili razlik v ultrastrukturi primarnih epitelnih celic MC.

Sekundarni povzetek: The Herald moth (Scoliopteryx libatrix) is a member of the family Erebidae in the superfamily Noctuidea. Two generations exist, the second one often overwinter in diapause in the entrance caves sections. The aim of the research was to find out how do the mass, the lipid and glycogen stores and water content change during overwintering, and if there exist diferences between the sexes. We also wanted to find out which changes in ultrastructure occur in the epithelial cells of the Malpighian tubules (MT) during this period of natural starvation. We performed optic and transmission electron microscopy, and adequate biochemical analyses in the study. It was found out that lipids are the main energy stores in S. libatryx during overeintering. The MTs consist of a monolayer epithelium, a basal lamina and individual muscle cells. The epithelium consists of primary epithelial cells attached to the basal lamina with hemidesmosomes. Between wrinkles of the basal plasma membranes there are mitochondria. In most cells nuclei are in their basal portion. Nuclei consist of heterochromatin, and are surrounded by an abundant rough endoplasmatic reticulum. The perinuclear portion is filled with spherites. Below the apical membrane there is the actin cortex containing a lot of mitochondria. The apical membrane is differentiated into numerous microvilli, which often contain mitochondria. Microvilli conspicuously enlarge the surface active in secretion into the lumen of the MTs. During overwintering, spherites gradually get emptied along with the appearance of autophagic structures, including residual bodies. No differences were found in the ultrastructure of the primary epithelial cells of MTs during overwintering.
